摘  要:目的:研究双孔钾通道TREK-1对体外培养的星形胶质细胞缺氧条件下活化增殖的影响。方法:离体培养大鼠大脑皮质星形胶质细胞,采用流式细胞术和BrdU掺入法检测其在缺氧条件下及给予TREK-1通道阻断剂干预后的活化增殖特点。结果:缺氧导致处于S期的星形胶质细胞明显增加,缺氧6h增加最明显;抑制TREK-1活性可加剧缺氧条件下星形胶质细胞的活化增殖。结论:星形胶质细胞的TREK-1活性与缺血缺氧状态下细胞的活化增殖密切相关。Objective: To investigate the effect of TREK 1 channel activity on the prolifera- ted astrocytes under hypoxia condition in vitro. Methods: Astrocytes from mice were cultured and exposed to hypoxia treatment. The proliferation of astrocytes and the effect of TREK-1 blockers on astrocytie proliferation under hypoxia condition were investigated by cytometry and BrdU incorporation. Results: The percentage of astrocytes in S phase was increased significantly under hypoxia condition as compared to that in the normal condition, which peaked at 6 h. The TREK-1 blockers promoted the proliferation of astrocytes under hypoxia condition. Conclusion: The TREK-1 channel activity is involved in the proliferation of astrocytes under hypoxia condi- tion.
