摘  要:目的研究大鼠脑梗死灶周围巨噬细胞游走抑制因子(MIF)的表达及变化,探讨MIF在脑梗死过程中的作用及意义。方法采用电凝法构建大鼠大脑中动脉闭塞模型,随机分为假手术组8只和脑梗死组48只,其中脑梗死组又分为4 h、12 h,1 d、3 d,1周、2周6个时间点,每个时间点8只,应用蛋白免疫印迹和荧光定量PCR技术检测脑梗死大鼠梗死灶周围MIF蛋白含量及其mRNA的表达水平。结果脑梗死组大鼠梗死灶周围MIF蛋白及mRNA表达在梗死后逐渐升高,梗死后4 h、12 h,1 d、3 d明显高于对照组(P<0.05)。大鼠梗死灶MIF蛋白含量与mNSS评分和MIF mRNA呈正相关(r=0.661,P=0.000;r=0.733,P=0.000)。结论大鼠脑梗死灶周围MIF蛋白及其mRNA表达水平呈先增高后恢复正常的变化规律,提示MIF在脑梗死病变过程中发挥重要作用。Objective To study the role of macrophage migration inhibitory factor(MIF) expres- sion in pericerebral infarction rats. Methods A rat right middle cerebral artery occlusion model was established by electrocoagulation. Fifty-six rats were randomly divided into sham operation group(n=8) and cerebral infarction group(n:48). The cerebral infarction group was further divided into 12 h group,1 d group,3 d group,1 week group and 2-week group,8 in each group. Ex- pression level of MIF protein and mRNA in pericerebral infarction rats was measured by Western blot and quantitative PCR,respectively. Results The expression level of MIF protein and mRNA was significantly higher in cerebral infarction group than in control group 4 and 12 h,1 and 3 d after cerebral infarction(P〈0.05). The MIF protein expression level was positively related with the MIF mRNA and mNSS score(r=0. 661,P=0. 000;r=0. 733,P=0. 000). Conclusion The expression level of MIF protein and mRNA increases and then retruns to normal in pericerebral infarction rats,indicating that MIF plays an important role in occurrence of cerebral infarction.
