摘 要:目的:观察n-6/n-3多不饱和脂肪酸(polyunsaturated fatty acid,PUFA)不同比例对乳腺癌细胞增殖能力、膜脂构成和膜功能的影响。方法:n-6/n-3PUFA不同比例处理MCF-7和MDA-MB-231乳腺癌细胞,气相色谱法分析细胞膜脂构成,共聚焦显微镜监测细胞膜流动性、膜电位和Ca2+跨膜内流。结果:单纯n-6和10∶1n-6/n-3PUFA组细胞膜较对照组含有更多的C18∶2和C20∶4,无n-3PUFA蓄积;但单纯n-3和1∶1n-6/n-3PUFA组细胞膜EPA含量显著增加,且未检测到C20∶4。另外,单纯n-6和10∶1n-6/n-3PUFA组可增加两种乳腺癌细胞的膜流动性,并诱导膜去极化和细胞外Ca2+内流;而单纯n-3和1∶1n-6/n-3PUFA组则降低细胞膜流动性,诱导细胞膜超极化改变和减缓细胞外Ca2+内流。结论:不同比例n-6/n-3PUFA明显改变MCF-7和MDA-MB-231乳腺癌细胞膜脂组成、膜流动性、超极化和Ca2+内流,这些改变可能参与其影响乳腺癌细胞生长的分子机制。Objective To investigate the effects of different ratios of n-6/n-3 polyunsaturated fatty acid (PUFA) on proliferation capacity, membrane lipid composition, and membrane function in MCF-7 (ER+) and MDA-MB-231 (ER-) human breast cancer cell lines. Methods MCF-7 (ER+) and MDA-MB-231 (ER-) human breast cancer cell lines were treated with different ratios of n-6/n-3 PUFA. Then the membrane lipid composition was analyzed by gas chromatography, and membrane fluidity, membrane potential, and Ca2+ influx were observed by confocal laser microscopy. Results As compared to those in control group, there were more C18:2 and C20:4 and no n-3 PUFA accumulation in plasma membrane in n-6 PUFA and 10:1 n-6/n-3 PUFA groups; in contrast, there were more EPA and no C20:4 accumulation in plasma membrane in n-3 PUFA and 1:1 n-6/n-3 PUFA groups. Additionally, the membrane fluidity, membrane depolarization, and Ca2+ influx were elevated in n-6 PUFA and 10:1 n-6/n-3 PUFA groups, whereas the opposites were found in n-3 PUFA and 1:1 n-6/n-3 PUFA groups. Conclusion Different ratios of n-6/n-3 PUFA can alter the membrane lipid composition, membrane fluidity, membrane hyperpolarization, and Ca2+ influx in MCF-7 and MDA-MB-231 breast cancer cells, which may be involved in the molecular mechanism of its effects on the growth of these cells.
