摘 要:目的研究亚致死剂量脉冲电场(pulse electric fields,PEFs)作用后HeLa细胞恶性生物学行为变化,探讨不可逆性电穿孔消融治疗是否有促进残留肿瘤细胞增殖、侵袭、转移的潜在可能。方法利用电场强度为500、750、1 000 V/cm的亚致死剂量PEFs分别作用于HeLa细胞,取正常野生株HeLa细胞作为对照。采取CCK8比色法检测细胞活性反映HeLa细胞增殖能力;基质胶黏附实验、Transwell小室体外侵袭实验和细胞迁移实验分别检测电场作用后HeLa细胞对基质的黏附能力、体外侵袭能力和细胞迁移能力的改变。结果不同场强亚致死剂量PEFs作用后12、24、48、72 h,HeLa细胞的增殖与对照组比较无显著改变。场强为500、750、1 000 V/cm的PEFs作用后,对HeLa细胞黏附抑制率分别为9.38%、30.21%和42.70%。对照组、500 V/cm组、750 V/cm组和1 000 V/cm组:体外侵袭穿透基质胶的细胞数分别为(104.17±4.59)、(101.82±5.76)、(78.39±6.18)、(67.91±5.94),其中750 V/cm组、1 000 V/cm组明显少于对照组(P<0.05);细胞划痕修复率分别为(53.46±8.07)%、(48.75±9.13)%、(36.49±7.36)%、(30.58±5.91)%,对照组显著高于750 V/cm组和1 000 V/cm组(P<0.05)。亚致死剂量PEFs对细胞的黏附、侵袭和迁移能力均呈现不同程度的抑制作用,其抑制效应和电场强度呈剂量依赖关系。结论体外实验中亚致死剂量PEFs对HeLa细胞的增殖无显著影响,但能够对HeLa细胞的侵袭和转移能力产生不同程度的抑制作用。提示不可逆性电穿孔在体外即使不能一次性彻底消融HeLa细胞,短期内也不会促进残留细胞的恶性生物学行为。Objective To investigate the malignant biological behavior of HeLa cells treated with sub- lethal dosage of pulse electric fields (PEFs) , and to explore the possibilities of the proliferation, metastasis and invasion of residual malignant cells promoted by irreversible electroporation ablation. Methods HeLa cells were treated with sublethal dosage of PEFs (500,750 and 1 000 V/cm, respectively) , and normal HeLa cells were used as control. The viability of HeLa cells was detected by cell counting kit-8 (CCK8). Matrigel adhe- sion experiment, Transwell chamber and scratch wound assay were used to detect the adhesive ability, invasion and migration of HeLa cells after treated with the sublethal dosages of PEFs, respectively. Results The prolif- eration of HeLa cells was the same as that of the control cells at 12, 24, 48 and 72 h after ablation by varied sublethal dosages of PEFs. With sublethal dosage of PEFs (500,750 and 1 000 V/cm), the rates of inhibition on HeLa cell adhesion were 9.38% , 30.21% and 42.70% , respectively. From the control group to the three experimental groups, the numbers of HeLa cells invaded through the matrigel were 104.17 ±4.59, 101.82 ±5.76, 78.39± 6.18 and 67.91 ±5.94, respectively, and the wound healing rates were (53.46± 8.07 ) % , (48.75± 9.13 ) % , (36.49 ±7.36) % and (30.58± 5.91 ) %, respectively. The number of HeLa cells inva- ded through the matrigel in the 750 V/cm group and 1 000 V/cm group was significantly less than that in thecontrol group (P 〈 0. 05 ), and the wound healing rate of the control group was significantly greater than those of the 750 V/cm group and 1 000 V/cm group. The sublethal dosage of PEFs could inhibit the adhesion, inva- sion and migration of HeLa cells in a dose-dependent manner. Conclusion Sublethal dosage of PEFs does not affect HeLa cell proliferation, but can inhibit HeLa cell invasion and metastasis in vitro. It suggests that even though irreversible electroporation can not completely ablate HeLa cells
