摘 要:目的:探讨Exendin-4对人肝癌细胞HepG2增殖的影响。方法:给予不同浓度(0.1、1.0、10.0、100.0nmol/L)的Exendin-4刺激HepG2,使用cell counting kit-8(CCK-8)试剂检测细胞增殖,流式细胞术检测细胞周期分布。结果:Exendin-4在0.1、1.0nmol/L水平对人肝癌细胞HepG2无明显促进增殖作用,在10.0、100.0nmol/L水平有明显促进增殖作用,差异具有统计学意义(P<0.05),呈现出浓度、时间依赖性地促进HepG2的增殖,并且随着Exendin-4水平的增加,HepG2G0/G1期细胞比例逐渐下降(P<0.05),S期及G2/M期细胞比例逐渐上升(P<0.05)。结论:Exendin-4在低浓度时对人肝癌细胞HepG2无明显促进增殖作用,但浓度增加到一定水平时具有促进增殖作用,且呈现时间依赖性促进H epG2的增殖效应。Objective: To investigate the effect of Exendin-4 on growth and proliferation of human hepatocellular carcinoma cells HepG2.Methods: HepG2 cells were cultured in vitro with Exendin-4 at different concentrations(0.1,1.0,10.0,100.0 nmol/L).Cell proliferation was detected by cell counting kit-8 reagents,and cell cycles were measured by flow cytometry.Results: No significant difference in cell proliferation was detected between control and Exendin-4 groups at the concentrations of 0.1 and 1.0 nmol/L,while difference in cell proliferation was significant between control and Exendin-4 groups at the concentrations of 10.0 and 100.0 nmol/L(P〈0.05).Exendin-4 promoted the proliferation of HepG2 in a dose and time-dependent manner.The cell ratio in G0-G1 phase decreased gradually(P〈0.05) and the ratio in S/G2-M phase was increased(P〈0.05).Conclusions:It is not obvious in promoting Proliferation of human hepatocellular carcinoma cells when Exendin-4 is at a low concentration.However,it has the effect of promoting Proliferation when the concentration increase to a certain level.Exendin-4 promoted the proliferation of HepG2 in a time-dependent manner.
