摘  要:环磷酸腺苷(cAMP)作为第二信使参与多种生理生化过程的调节,是人体内重要的生理活性物质。本文采用水浸提和6000Da超滤膜过滤方法对大枣进行预处理,从5种树脂中选型最合适的树脂,并对提取工艺进行确定。结果表明,大枣在小颗粒状(0.3cm)捣碎状态下,60℃水浸提12h,cAMP提取效果最好。通过考察SD5、SD8、D01、D05和D13五种型号离子交换树脂,发现D13型树脂提取大枣中cAMP效果最佳。选用的洗脱剂为0.05mol/LHCl,流速为1.5mL/min。得到产品产率为65.0%,其中cAMP含量37.5%。Cyclic adenosine-3', 5'-monophosphate (cAMP) is one of the most important physiological active substances as the second messenger for regulating physiological and biochemical processes. Water extraction and 6 000 Da ultrafiltration method were used for pretreatment. The most suitable one was selected from 5 types of resins and the extraction process was optimized. Results showed that the optimal pretreatment condition for jujubes is that 12 hours water extraction at 60℃ with small granular (0.3 cm) broken state. By comparing SD5, SD8, DO1, DO5 and D13 ion exchange resins, D13 was found to be the most suitable one for cAMP extraction. 0.05 mol/L HC1 were selected as eluting agent at the flow rate of 1.5 mL/min. The product yield was 65% while the cAMP concentration was 37.5%.
