摘 要:目的探讨乳腺癌组织中雌激素受体(ER)β1、ERβ2基因和蛋白的表达及其临床意义。方法应用Western印迹法检测乳腺癌组织、癌旁组织及正常组织中ERβ1和ERβ2蛋白的表达情况,荧光定量聚合酶链反应(PCR)检测ERβ1、ERβ2mRNA的表达情况。结果乳腺癌组织和癌旁组织中的ERβ1mRNA、ERβ2mRNA检出率与蛋白检出率相同。乳腺癌组织中的ERβ1蛋白检出率显著低于癌旁组织(P=0.029)和正常组织(P=0.029);乳腺癌组织中的ERβ1蛋白表达量为47.06±8.94,显著低于癌旁组织中的55.33±9.59及正常组织中的54.47±9.65(P=0.009及0.019)。乳腺癌组织中的ERβ1mRNA检出率显著低于癌旁组织(P=0.029);乳腺癌组织中的ERβ1mRNA表达量为0.026±0.014,显著低于癌旁组织中的0.041±0.020(P=0.015)。乳腺癌组织中,ERβ1mRNA与蛋白表达量的相关系数为0.635(P=0.008),ERβ2mRNA与蛋白表达量的相关系数为0.714(P=0.004);癌旁组织中,ERβ1mRNA与蛋白表达量的相关系数为0.506(P=0.012),ERβ2mRNA与蛋白表达量的相关系数为0.926(P=0.000),ERβ1mRNA、ERβ2mRNA与蛋白水平之间存在相关性。结论全长野生型ERβ1可能是避免乳腺癌发生的保护性因子,ERβ1有助对乳腺癌预后的预测。Objective To explore the expression of estrogen receptor (ER) β1 and ERβ2 mRNA and protein in breast cancer and its clinical significance. Methods Western blotting analysis was carried out to measure the protein expression of ERβ1 and ERβ2 in breast cancer tissue, adjacent tissue and normal tissue. Fluorescence quantitative PCR was applied for detecting the mRNA expression of ERβ1 and ERβ2. Results The positive rate of ERβ1 mRNA/ERβ2 mRNA in both breast cancer tissue and adjacent tissue was the same as that of ERβ1 protein/ERβ2 protein. The positive rate of ERβ1 protein in breast cancer tissue was significantly lower than that in adjacent (P=0.029) and normal tissues (P=0.029). The relative protein expression of ERβ1 in breast cancer tissue (47.06±8.94) was significantly lower than that in adjacent tissue (55.33±9.59) or in normal tissue (54.47±9.65, P=0.009 and 0.019). The positive rate of ERβ1 mRNA in breast cancer tissue was significantly lower than that in adjacent tissue (P=0.029). The mRNA level of ERβ1 in breast tissue (0.026±0.014) was significantly lower than that in adjacent tissue (0.041±0.020, P=0.015). In breast cancer tissue, the correlation coefficient of ERβ1 mRNA and protein expression was 0.635 (P=0.008), and the correlation coefficient of ERβ2 mRNA and protein expression was 0.714 (P=0.004). In adjacent tissue, the correlation coefficient of ERβ1 mRNA and protein expression was 0.506 (P=0.012), and the correlation coefficient of ERβ2 mRNA and protein expression was 0.926 (P=0.000). There was a positive correlation between mRNA and protein levels of both ERβ1 and ERβ2. Conclusion Total length wildtype ERβ1 may be a preventer of breast cancer and be helpful in predicting the prognosis of breast cancer.
