摘  要:目的观察α-硫辛酸(ALA)对高脂饮食诱导肥胖大鼠血清胱抑素C(Cys C)水平及肾脏病理的影响。方法 Wistar大鼠50只,随机分为普通饲料组(正常对照组,n=10)、高脂饮食组(n=40)。成功诱导的肥胖大鼠再随机分为HFD组(n=16)和HFD+ALA组(n=16),HFD+ALA组给予为期2周的ALA[30 mg/(kg.d)]腹腔注射。测定大鼠血清空腹血糖(FBG)、空腹胰岛素(FIns)、Cys C、血脂等指标;HE、PAS染色观察各组大鼠肾脏的病理变化。结果高脂饮食成功建立肥胖大鼠模型32只;与正常对照组相比,HFD组FIns、总胆固醇(TC)、三酰甘油(TG)等明显升高(P<0.05),Cys C浓度[(0.16±0.04)mg/L]亦高于正常对照组[(0.06±0.03)mg/L]。ALA干预使肥胖大鼠体重降低,不同程度地降低游离脂肪酸、FIns、胰岛素抵抗指数、TC、TG;血清Cys C水平下降[(0.04±0.03)mg/L]。结论高脂饮食诱导的肥胖大鼠Cys C水平升高,ALA明显改善肥胖大鼠的胰岛素抵抗状态,降低肥胖引起的肾损伤。Objective To evaluate the effects of alpha - lipoie acid (ALA) on the function and morphology change of kidney in high - fat diet (HFD) - induced obese rats. Methods Fifty Whistar rats were randomly divided into control group, HFD group and HFD + ALA group. Subjects in HFD + ALA group were treated with ALA (30 mg/kg) in- traperitoneally for 2 weeks. The concentrations of serum FBG, Fins, Cys C and lipids were determined. The pathological change in kidney were observed with HE and PAS stain. Results Thirty - two obese rats were obtained with HFD. Com- pared with control group, significantly higher Fins, TC, TG and Cys C were revealed in HFD group (P 〈 0. 05). Moreo- ver the ALA intervention significantly inhibited the HFD - induced elevation in weight, Cys C, NEFA, Fins, IRI, TC and TG. Conclusion ALA can alleviate the HFD -induced IRI and renal injury in rats.
