摘 要:目的通过检测CD256/CD257在肾癌组织中的表达特点,分析CD256/CD257表达与肾癌临床病理分期、分级以及病理分型的关系。方法依据2002年AJCC肾癌TNM分期标准对90例肾癌组织进行病理分析。采用免疫组化技术(S-P法)和荧光定量PCR技术检测CD256、CD257在组织病理切片的表达情况。与临床病理特征结合进行统计分析。结果研究发现CD256、CD257在肾癌的表达与肾癌分期和分级没有显著相关性,但是CD256在肾透明细胞癌肿瘤组织中有高表达,在肾嫌色细胞癌肿瘤组织中无表达,而在两类肿瘤的癌旁组织中均为弱表达,表达于肿瘤细胞或肾小管上皮细胞的胞质/胞膜;CD257在肾透明细胞癌肿瘤组织中不表达,在肾嫌色细胞癌肿瘤细胞中等至高表达,而在两类肿瘤的癌旁组织中均为中等至高表达,表达于肿瘤细胞或肾小管上皮细胞的胞质/胞膜。CD256/CD257mRNA水平与蛋白水平一致。CD256、CD257在肾透明细胞癌和肾嫌色细胞癌呈现的差异性表达具有统计学意义(P<0.05)。结论 CD256、CD257在肾透明细胞癌和肾嫌色细胞癌的差异性表达可能与不同类型肾细胞癌的发生与分化有着紧密联系,值得深入研究。Objective To investigate the expression characteristic of CD 256/CD257 on renal cell carcinoma and the correlation with clinical pathologies. Methods 90 renal carcinoma eases were selected, and the excisional tissue sections were analyzed by pathologists according to 2002 AJCC Renal Carcinoma TNM staging criteria, and were detected of CD256 and CD257 by immunohistochemical assay(S-P). Combined analysis was taken with clinical pathological features. Results The present results showed that there were no significant correlation of CD 256/CD257 with clinical pathological stages and types. However, CD256 high expressed on clear cell renal cell carcinoma (ccRCC) , but not on chromophobe carcinoma , and weakly expressed on the para-ccRCC carcinoma tissues. CD257 did not expressed on ceRCC tissues, while moderately or high expressed on chromophube carcinoma, and also moderately or high expressed on para-chromophobe carcinoma tissues. CD256 and CD257 all stained in tumor cells and cytoplasm or membrane of renal tubular epithelial cells . Real-time PCR results showed that CD256/CD257 mRNA levels were coincident with the results of IHC. These differential expression rate of CD 256/CD257 on ccRCC and chromophobe carcinomas were significant ( P 〈 0. 05 ). Conclusions The significant differential expression of CD256/CD257 may closely correlated with development and differentiation of renal cell carcinomas , which worthy deeply studying.
