ORIGINS OF THE CENTRAL AFFERNTS TO DMN AND THEIR PEPTIDERGIC TRANSMITTERS

摘  要:用辣根过氧化物酶(HRP)逆行结合免疫组化定位的双标法,观察大鼠迷走背核(DMN)的中枢传入来源和位于这些神经通路的肽能递质。(1)将麦芽凝集素结合HRP(WGA-HRP)注入DMN脑区后,见HRP逆行标记细胞位于:①脑干:孤束核、三叉神经脊束核、小细胞网状核、外侧网状核、中缝隐核、中缝苍白核、臂旁核、被盖背核及蓝斑;②边缘前脑:下丘脑室旁核(PVH)、杏仁中央核、终纹床核和岛叶皮质(IC)。(2)部分切片继续进行免疫组化反应,观察HRP和肽共存的双标细胞。在PVH的尾侧部见辣根过氧化物酶-催产素(HRP-OT)/加压素(VP)/生长抑素(SOM)/P物质(SP)双标细胞。IC有少量散在分布的HRP-SOM/SP双标细胞。结果提示:(1)DMN接受各级中枢的传入,其中起自PVH和IC部分投射中含SOM、SP和/或OT、VP免疫反应性物质;(2)边缘叶皮质对DMN的调节可能至少通过两条途径:①起自IC到DMN的直接通路;②以皮质下核和/或网状结构等为中介的间接通路。A double-labeling method that combines the retrograde transport of horseradish per-oxidase (HRP) with immunohistochemical procedure was used to demonstrate the origins of the central afferents to dorsal motor nucleus of vagus (DMN)and to identify the pe-ptidergic transmitters in these neuropathways in rats.(1) After injecting wheat germ ag-glutinin conjugated HRP (WGA-HRP)into the neural area of DMN, the HRP labeled neurons were found in the following regions:(1) Brainstem: solitary nucleus (n.), spinal tract n. of trigeminal nerve, parvocellular reticular n., lateral reticular n., obscure raphe n., pallidus raphe n., parabrachial n., dorsal tegmental n., and locus ceruleus: (2) Limbic forebrain: paraventricular n. of hypothalamus (PVH), central amygdaloid n., bed n. of stria terminalis and insular cortex (IC). The labeled cells of IC are medium size pyramidal cells which are densly localized in the fifth layer of an area meassured approximately 1×2mm, rostralateral to anterior commissure and ventral to rhinal fissure. (2) Some sections, after the stabilization of retrograde HRP, were again stained immunohist-ochemically to show the coexistence of HRp and peptide in the same neuron. HRP-OT (oxytocin)/VP (vasopressin)/SOM (somatostatin)/SP (substance P) double-labeled cells Were located primarily in the caudal part of PVH. A few HRP-SOM/SP cells were dispersed in IC. The results suggest that: (1) DMN receives afferent projections from various brain regions, and some of the projections from PVH and IC are SOM, SP and/or VP peptider-gic; (2) The limbic cortex controls DMN probably through at least two pathways: one originates from IC and goes directly to DMN; another projects indirectly through the intermediates of subcortical nuclei and/or reticular formation. This pattern of regulation appears similar to that of somatomotor regulation through pyramidal and extropyramidal connections.
