摘  要:目的探讨转化生长因子-β1(TGF—β1)诱导胃癌细胞的凋亡作用及机制。方法利用TGF-β1诱导胃癌细胞SGC7901凋亡,小干扰RNA(siRNA)封闭半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-8的表达,Westernblot检测诱导及封闭前后各Caspases表达随时间的变化,流式细胞术分析细胞的凋亡率。结果TGF—β1作用后,各组细胞的凋亡率明显增加,0、24、48h凋亡率分别为1.13%、14.88%、37.32%;Caspase-812h后激活,Caspase-924h后激活;白RNA干扰(RNAi)后Caspase-8表达明显下降,24h后Caspase-8表达量为对照组的11.2%,48~72h未测到表达,96h后Caspase-8表达重新出现,为对照组的9.2%。结论TGF—B1通过Caspase途径诱导胃癌细胞发生凋亡,由Caspase-8开始启动;抑制Caspase-8表达可以在一定程度上阻断TGF—β1/Caspases信号的转导,进而抑制细胞凋亡。Objective To investigate the effects of transforming growth factor-β1 ( TGF-β1 ) in- duced apoptosis. Methods Apoptosis proportion of human gastric SGC7901 carcinoma cells induced by TGF-β1 was tested by flow cytometry, blocking Caspase-8 by RNA interference (RNAi) , the expression of Caspase-8 was analyzed by Western blotting. Results The apoptosis rate of SGC7901 cells obviously increased with the treatment of TGF-β1, an active fragment of caspase-8 started to occur 12 h after TGF-β1 treatment, whereas the caspase-9 active fragment occurred 24 h after TGF-β1 treatment. After RNAi a- gainst Caspase-8, the expression of Caspase-8 was decreased, 11.2% of control group at 24 h, and then can not be detected at 48 h, 72 h, re-emergence at 96 h, 9.2% of control group. Conclusion TGF-β1 induced the apoptosis of SGC7901 cells via Caspases pathway, caspase-8 may be the first easpase activa- ted;Suppression of caspase-8 by RNAi can effectively block the TGF-β1/Caspases signal pathway, and subsequently inhibits the apoptosis of SGC7901 cells.
