摘  要:目的:通过研究不同时间点金欣口服液对呼吸道合胞病毒(RSV)感染模型肿瘤坏死因子-α(TNF-α)表达的影响,进一步阐明金欣口服液治疗RSV肺炎的作用机制。方法:体外实验:RSV感染体外培养的RAW264.7细胞,金欣口服液合药血清进行干预,24h后收集细胞上清,ELISA法检测各组TNF-α含量;体内实验:RSV滴鼻感染BALB/c小鼠,金欣口服液不同剂量及给药方式进行干预,并于首次滴鼻后24、72、144h取各组小鼠支气管肺泡灌洗液,ELISA法检测TNF-α表达及金欣口服液的干预效果。结果:RSV感染RAW264.7细胞及BALB/c小鼠后24h,TNF-α表达明显升高(P<0.01),而金欣口服液组较RSV感染组TNF-α表达明显下调(P<0.01);72h后,RSV感染小鼠体内TNF-α表达虽较正常组高(P<0.05),但较24h组明显下降(P<0.01),而金欣口服液等效剂量预防及治疗组TNF-α表达较RSV感染组均明显升高(P<0.01);144h后,RSV感染小鼠体内TNF-α表达较正常组无差异(P>0.05),而金欣口服液高剂量及等效剂量治疗组TNF-α表达均较RSV感染组明显升高(P<0.05)。结论:金欣口服液能显著下调RSV感染初期TNF-α的高表达,且随着感染时间的延长,可适当上调TNF-α的表达,表明金欣口服液对RSV感染后体内TNF-α的表达具有一定的双向调节趋势,这可能是其治疗RSV肺炎的作用机制之一。Objective:To investigate the influence of Jinxin oral liquid on the expression of TNF-α in RSV infected models.Methods:In vitro experiment:RAW264.7 cell line was infected with RSV,and treated with the drug serum of Jinxin oral liquid.24 hours later,the supernatant was collected and the level of TNF-α was measured.In vivo experiment:BALB/c mice were inoculated with RSV,and treated with Jinxin oral liquid of different dosage and delivery method.After 24,72 and 144 hours of the first inoculated with RSV,the mice were killed and the bronchoalveolar lavage samples were collected,then the level of TNF-α was measured.Results:In vitro and vivo experiments,it demonstrated that after 24 hours of infection with RSV,the expression of TNF-α was up-regulated significantly compared with that in the control group(P0.01).The expressions of TNF-α in Jinxin oral liquid groups were typically down-regulated compared with that in RSV infected groups(P0.01).After 72 hours of inoculation with RSV,although the expression of TNF-α in model group was higher than that in the control group(P0.05),it was much more lower than that in the model group of 24 hours(P0.01).The treating and pre-treating groups with equivalent dosage of Jinxin oral liquid significantly up-regulated the expression of TNF-α(P0.01).After 144 hours of inoculation with RSV,there was no difference between the model and control group for the secretion of TNF-α(P0.05),but the treating group with high and equivalent dosage of Jinxin oral liquid up-regulated the expression of TNF-α typically(P0.05).Conclusion:Jinxin oral liquid can down-regulate the expression of TNF-α for the cell and animal models at the early stage of infection with RSV.With the extension of RSV infection,Jinxin oral liquid can up-regulate the expression of TNF-α.All of these demonstrate that Jinxin oral liquid can regulate the expression of TNF-α in RSV infected models with the two-way adjustment,which may be one of the mechanism for Jinxin oral liquid in
