摘 要:目的观察茶多酚(TP)对实验性肥胖大鼠脂肪组织PPAR-γ表达的影响。方法通过高糖高脂饮食诱导建立实验性肥胖SD大鼠模型。给予TP干预10周后测定各组大鼠血糖、血脂、胰岛素、FFA水平,计算胰岛素抵抗指数(HOMA-IR)。分别用荧光定量PCR法、Western blot、免疫组化染色法检测各组大鼠脂肪组织PPAR-γ在基因及蛋白水平的表达。结果 TP组大鼠FPG、TG、TC、LDL-C、FFA、HOMA-IR均较肥胖大鼠对照组明显下降(P<0.05),TP组脂肪组织PPAR-γ在基因及蛋白水平的表达较肥胖大鼠对照组均有增加(P<0.05)。结论 TP可调控大鼠脂肪组织PPAR-γ的表达,并显著改善肥胖大鼠糖脂代谢,有望成为临床治疗肥胖及MS的有效药物。Objective To observe the effect of tea polyphenol on PPAR-7 expression in adipose tissue of experimental fatty rats. Methods The model of experimental fatty rat induced by high glucose and high {at diet was established. Ten weeks after intervention of tea polyphenol, the levels of blood glucose, blood fat, fasting insulin and free fatty acid were detected, and then the insulin resistance index was calculated. The expression of PPAR-7 in adipose tissue was detected by fluorescent quantitative polymerase chain reaction assay, Western blot and immunohistochemistry staining assay. Results The levels of fasting blood glucose [ (6.3 ±0. 7) vs (7. 1± 0. 8) rnmol/L], triglyceride [-(2.01±0. 99) vs (3.98±1.69) mmol/L], cholesterol [(3.31±1.96) vs (6. 98± 2. 88) rnrnol/L], LDL-C [(1.24±0. 39) vs (1. 985±0. 56) mmol/L], free fatty acid [(153. 24-31.4) vs (259. 5 ±56.9) mol/L], and HOMA-IR (7. 47 ± 1.35) vs (10.25 ± 1.98) were significantly lower in the tea polyphenol group than in the control group (P〈0.05). Compared with the control group, the mRNA and protein expressions of PPAR-y in adipose tissue of the tea polyphenol group were upregulated (P〈0. 05). Conclusion Tea polyphenol could modulate the expression of PPAR-7 in adipose tissue and improve the glucolipid metabolism. It may become an effective drug to treat metabolic syndrome.
