摘　　要：在采用PCR -SSCP(PolymeraseChainReaction -SingleStrandConformationPolymorphism ,聚合酶链式反应 -单链构象多态性 )分析技术对 8例骨关节病患者软骨细胞线粒体DNA进行突变分析的过程中 ,针对区带多而杂乱的情况 ,改变变性剂成份和电泳电压 ,从而有效提高了检出灵敏度 ,同时极大地节省了时间。PCR-SSCP is one of the extensively applied methods in detecting gene mutations. In this paper, it was used in mutation detection of ATP enzyme encoding sequence on mitochondrial DNA (mt DNA) of chondrocytes from 8 patients of osteoarthrosis. Our methodically improving strategies of using SDS to assist denaturing of DNA fragments and electrophoresis voltage high to 18V/cm were here introduced.
