摘  要:目的 :探讨卵巢癌多药耐药 (MDR)相关药物的体外敏感性与 MDR1基因表达的关系 ,为卵巢癌合理化疗提供理论依据。方法 :采用四甲基偶氮唑蓝 (MTT)比色法体外药物敏感 (药敏 )试验及逆转录—聚合酶链反应 (RT- PCR)技术 ,检测 37例卵巢癌对 MDR相关药物的敏感性 ,及其 MDR1基因表达情况。结果 :卵巢癌 MDR1基因表达阳性率为30 % ,与 MDR相关药物耐药有关 ,与非 MDR相关药物耐药无关。结论 :MDR1基因表达是卵巢癌抗 MDR相关药物的主要原因。Objective:To study the relationship between in vitro chemosensitivity of ovarian cancer (OC) to MDR related drugs and expression of MDR1 gene.Methods:Sensitivity of OC to clinical common anticancer agents was examined by means of MTT assay in vitro.Reverse transcription-polymerase chain reaction(RT-PCR)assay was used for dection of MDR1 gene expression in 37 OC.Results:MDR1 gene expression was detected in 30% of OC analysed.The level of MDR1 mRNA was not correlated with non MDR related drugs,but correlated with MDR related drugs.Condusion:These results support the idea that the increased expression of MDR1 gene is a major cause of resistance to MDR related drugs.
