摘  要:目的:研究河豚毒素(tetrodotoxin,TTX)与吗啡联合使用对吗啡依赖性及镇痛耐受作用的影响。方法:采用吗啡依赖性小鼠模型及热板实验,评价单独使用吗啡(2.5 ml.kg-1)以及联合给药(吗啡2.5 mg·kg-1+TTX 0.5μg·kg-1、吗啡2.5 mg·kg-1+TTX 1.0μg·kg-1)对纳洛酮催促戒断症状及镇痛耐受性的影响。结果:联合用药可以抑制吗啡依赖性小鼠戒断后体重的丢失,明显抑制吗啡依赖小鼠纳洛酮催促后的跳跃反应,抑制小鼠热板实验的潜伏期时间的增加。结论:TTX与吗啡联合用药可以抑制吗啡的依赖性与镇痛耐受作用的产生。Obje.ctjve:To study the effect of tetrodotoxin (TTX) combined with morphine on the dependence and analgesic tolerance acting of morphine. Methods: The mouse model of morphine dependency and hot plate experiment were used in this research to investigate the effects of morphine alone( morphine 2.5 ml ·kg^-1 ) and morphine combined with TTX ( morphine 2.5 mg ·kg^-1+ TTX 0. 5μg·kg^-1, morphine 2.5 mg ·kg^-1 + TTX 1.0 μg·kg^-1) on the withdrawal symptoms induced by naloxone and the analgesic tolerance of morphine. Results: Administration of morphine combined with TYX could inhibit the weight loose of mice, significantly inhibit the jumping effect after the withdrawal experiment induced by naloxone and reduce the increase of the response latency in the hot- plate experiment. Conclusion: Administration of morphine combined with TFX can inhibit the effect of dependence and analgesic tolerance acting of morphine.
