盐酸多西环素联合碘油对兔VX2肝瘤凋亡、增殖及肝功能的影响研究

摘　　要：目的:研究盐酸多西环素(DOXY)联合碘油对兔VX2肝瘤凋亡、增殖及肝功能的影响。方法:取兔建立兔VX2肝瘤模型,2周后,随机分为A(生理盐水)组、B(碘油0.3mL.kg-1)组、C(阿霉素2mg·kg-1+碘油0.3mL.kg-1)组和D(DOXY3mg.kg-1+碘油0.3mL·kg-1)组,每组8只,经股动脉-肝动脉插管给予相应药物。测定治疗前1d及治疗后4、7d各组血清中天冬氨酸氨基转移酶(AST)、丙氨酸转氨酶(ALT)含量,以及治疗后7d肿瘤细胞凋亡指数和半胱氨酸天冬氨酸蛋白酶3(Caspase-3)表达、残存肿瘤增殖细胞的增殖指数。结果:与A组比较,B、C、D组治疗后4dAST、ALT含量均明显升高(P<0.05),治疗后7dAST、ALT含量逐渐下降,但3个组的AST含量仍明显偏高(P<0.05),C组ALT含量明显偏高(P<0.05);3个组凋亡指数和Caspase-3表达均明显升高(P<0.05),增殖指数明显降低(P<0.05)。与B组比较,C组治疗后4、7dAST、ALT含量均明显升高(P<0.05);D组凋亡指数和Caspase-3表达均明显升高(P<0.05),增殖指数明显降低(P<0.05)。结论:DOXY联合碘油对肝功能无明显损害,可加速肿瘤细胞凋亡,降低其增殖能力。DOXY促进肿瘤细胞凋亡可能与其上调Caspase-3表达有关。OBJECTIVE：To investigate the effect of doxycycline hydrochloride（DOXY） and lipiodol on the apoptosis and proliferation of VX2 hepatic tumor and liver function of rabbits.METHODS：VX2 implanted hepatic tumor model was established and 2 weeks later they were randomly divided into group A（normal saline）,group B（lipiodol 0.3 mL·kg-1）,group C（adriamycin 2 mg·kg-1＋lipiodol 0.3 mL·kg-1） and group D（DOXY 3 mg·kg-1＋lipiodol 0.3 mL·kg-1） via catheter inserted into the arteria cruralis and hepatic artery of rabbit.The levels of AST and ALT in serum were respectively tested at 1 day before treatment,and 4,7 days after treatment.The apoptosis index（AI） of tumor cells and the expressions of Caspase-3 were measured,and proliferation index（PI） of proliferating cells was also detected.RESULTS：Compared with group A,the contents of AST and ALT increased significantly in group B,C and D 4 days after treatment（P0.05）;7 days after treatment,the contents of AST and ALT tended to decrease,but the contents of AST were still higher in 3 groups（P0.05）,and the content of ALT higher in group C（P0.05）;AI and Caspase-3 expression of tumors in 3 groups increased significantly（P0.05）,and PI of tumor decreased significantly（P0.05）.Compared with group B,AST and ALT contents increased significantly in group C 4 and 7 days after treatment（P0.05）;AI and Caspase-3 expression of group D increased significantly（P0.05）,while PI decreased significantly（P0.05）.CONCLUSION：DOXY and lipiodol have nearly no significant damage on liver function,and can promote the apoptosis of tumor cells and reduce proliferation of it.DOXY promote the apoptosis of tumor cells which may be associated with the up-regulation of Caspase-3 expression.
