摘  要:目的:考察山楂叶总黄酮的平衡溶解度、油水分配系数等理化常数,研究温度、pH值等对其水溶液稳定性的影响。方法:测定山楂叶总黄酮在不同介质中的平衡溶解度与不同pH条件下的表观油/水分配系数(Papp),以10h后的药物含量变化率为指标考察介质、温度、pH对其稳定性的影响。结果:山楂叶总黄酮在蒸馏水、肠溶液、0.1%盐酸溶液、甲醇、乙醇、pH5.0 PBS、pH 6.5 PBS、pH 7.0 PBS、pH 7.4 PBS、pH 9.0 PBS中的平衡溶解度分别为2.81、5.07、1.62、60.76、31.57、3.78、4.54、8.74、9.24、9.15mg/mL;山楂叶总黄酮在pH 3.0、4.0、5.0、6.0、7.0、8.0条件下的油/水分配系数,分别为0.256、0.332、0.371、0.385、0.235、0.234。结论:山楂叶总黄酮的Papp值均随着pH增大而增大,在pH 6.0与pH 7.0间发生突变,pH 6.0时达最大值,之后随着pH增大而减小;较高pH条件对山楂叶总黄酮水溶液稳定性影响较大。Objective:To investigate the physicochemical constants just like equilibrium solubility and apparent parti- tion coefficients( papp), and study the effects of temperature and pH value on the stability of hawthorn leaves flavonids. Method:The equilibrimn solubility of hawthorn leaves flavonids in various medium and the papp of hawthorn leaves fla- vonids under different pH conditions can be determined by UV - Vis, the effects of temperature and pH value on the sta- bility of solution can be investigated by taking change ratios of drug content in lOh as detection index. Result:It can be known the equilibrium solubility of hawthorn leaves flavonids in distilled water, intestinal solution, O. 1% hydrochloric acid, methanol, ethanol, PBS with pH value 5.0, PBS with pH 6.5, PBS with pH 7.0, PBS with pH 7.4, PBS with pH 9.0 were 2.81,5.07,1.62,60.76,31.57,3.78,4.54,8.74,9.24,9.15 mg/mL, respectively; and its oil/water partition co- effieient in pH 3.0,4.0,5.0,6.0,7.0,8.0 were 0. 256,0. 332,0. 371,0. 385,0. 235,0. 234 ,respectively. Conclusion: The papp of hawthorn leaves flavonids was increased by the pH value, at pH 6.0,with the maximum, then decreased with the increasing pH value ; the stability of hawthorn leaves flavonids solution was apprantly inflused by high pH value ( pH 9.0).
