摘 要:通过对Bacillus subtilis E1R-j产脂肽物质的发酵条件的优化来提高脂肽物质产量,从而为工业化生产奠定基础。在Landy培养基中,采用单因素试验法确定对E1R-j菌株产脂肽物质的影响因素及水平,结合响应面法确定最佳发酵条件。结果表明,温度40℃、培养基初始pH 9.0、接种量3.0%、装液量50mL、转速200r/min为最佳发酵条件。在最佳发酵条件下培养,粗脂肽物质的质量浓度达1.39g/L,是原发酵条件下质量浓度0.55g/L的2倍多。优化后的培养条件提高了B.subtilis E1R-j产粗脂肽物质的量,表明响应面法是一种快速、有效、简单的优化方法。In order to lay the groundwork for industrialized production,the yield of lipopeptide produced by Bacillus subtilis E1R-j was improved by optimizing its fermentation conditions in Landy medium.In Landy medium,the factors and its levels influencing the production of lipopeptide produced by E1R-j strain,was determined by using single-factor test,and the optimization of fermentation conditions was approached by using RSM.The optimum fermentation conditions were as follows: temperature was 40 ℃,pH in Landy medium was 9.0,inoculation was 3.0%,medium volume was 50 mL,rotary speed was 200 r/min.Under the new condition,the mass concentration of crude lipopeptide was 1.39 g/L,which was more than twice as much of before(0.55 g/L).The largely improvement of the yield of lipopeptide produced by B.subtilis E1R-j suggest that RSM is a kind of very rapid,valuable and simple method.
