胶原酶A与大肠癌侵袭转移性的关系研究

摘　　要：目的　揭示胶原酶A (MMP 2 )与大肠癌分期、临床表现的关系 ,探讨以胶原酶A在肿瘤组织中的含量及活性比值来判定大肠癌侵袭转移的可能性及方法。方法　采用明胶酶谱法检测 3 2例大肠癌组织和 17例癌旁组织及 7例外伤或尸检的正常大肠组织中MMP 2的酶原和酶含量 ,并计算其活性比值。结果　MMP 2含量及活性比值在癌、癌旁及正常大肠组织中存在显著性差异 (P <0 .0 1) ,且与大肠癌Duke′s分期 (P <0 .0 1)、患者年龄 (P <0 .0 5 )相关 ,而与肿瘤组织分型、肿瘤部位、患者的性别无关。结论　MMP 2与大肠癌的发生发展 ,特别是与肿瘤的侵袭转移关系密切 ,有望成为判断肿瘤侵袭转移的有效的新指标 ;MMP 2表达不同可能是不同年龄患者大肠癌生物学行为差异的原因之一 ;明胶酶谱法是检测MMP 2的有效方法。Objective To explore the relationship between colleganase A and colorectal cancer Dukes' classification and clinical representations.Methods Gellatinase zymograpy was used to measure the content of colleganase A in 32 cases of colorectal cancer,17 cases of adjacent tissues and 7 cases of normal colorectal tissue,their association with Duke′s classification,sex,locality,age and tissue type was analyzed.Results The contents of colleganase A and the proportion of the active form were evidently different among colorectal cancer,adjacent tissue and normal tissue( P <0.01).They were correlated with Duke′s classification( P <0.01) and age( P <0.05).Conclusion The results suggest that the colleganase A may not only be correlated with occurrence and evolution of colorectal cancer,but with invasion and metastasis of colorectal cancer;The expression levels of the colleganase A in colorectal cancer may be correlated with the biological behavior of colorectal tumor changed with the age of patient;Gelatinase zymography is the effective method to measure the contents of collagenase A in tumor tissue.
