摘 要:目的 研究抑癌基因p16和Rb与肺癌生物学特征和临床进展的关系。方法 采用免疫组织化学和Southern杂交的方法对 68例肺癌进行了抑癌基因 p16和Rb的检测。 结果 肺癌 p16蛋白总丢失率为 48.5 % ,且主要发生于非小细胞肺癌病例 ,与淋巴结转移、临床病理分期有关。Southern杂交发现 p16基因缺失的频率为 17.6% ,缺失多发生于p16蛋白阴性的病例。肺癌Rb蛋白丢失率为 2 9.4% ,主要发生于小细胞肺癌病例。Southern杂交发现 2 3 .5 %的肺癌有Rb基因异常。结论 p16基因和Rb基因缺失是该基因失活的重要原因 ,是肺癌发生、发展中重要的分子事件 ,与肺癌的临床进展密切相关。Objective To study the relationship between p16 and Rb genes and primary lung cancers.Methods p16 and Rb gene in 68 cases of lung cancer were observed by immunohistochemistry and southern blot.Results The inactivation rate of p16 gene in lung cancer was 48.5% by immunohistochemistry,it related to histological type,lymph node metastasis and progressive stage.The deletion rate of p16 gene in lung cancer was 17.6% by southern blot,it presented mostly in the cases with negative p16 protein.The inactivation rate of Rb gene in lung cancer was 29.4% by immunohistochemistry and 23.5% by southern blot,and only related to histological type.The inverse correlation of p16 and Rb expression in lung cancer was confirmed.Conclusion The inactivation of p16 or Rb gene is an important molecular event in lung cancer and related to clinical progression of lung cancer.
