摘 要:目的探讨浙江地区肺腺癌患者中EGFR和K—ras基因的突变情况。方法微分离富集肿瘤细胞,采用QIAGENDNA提取试剂盒提取基因组DNA,通过PCR扩增及DNA测序技术(Sanger测序法)分析肺腺癌患者EGFR和K—ras基因突变情况。结果54例肺腺癌患者中有20例(37.0%]存在EGFR基因突变。其中外显子19突变14例(25.9%);外显子21突变6例(11.1%)。未检测到外显子18和20突变。K—ras突变1例(2%)。外显子19男性患者的突变率为29-2%(7/24)。女性为23.3%(7/30):两者差异无统计学意义[P〉0.05);外显子21男性患者的突变率为4%(1/24)。女性为16-7%(5/30),两者差异无统计学意义(P〉0.05)。结论浙江地区肺腺癌患者中,男性和女性的突变率无明显差异;EGFR基因的突变以外显子19的缺失突变最常见;K—ras突变较为罕见。Objective To investigate EGFR and K-ras gene mutations in patients with lung adenocarcinoma in Zhejiang province. Methods Tissue specimens were collected from 54 patients with lung adenocarcinoma in Zhejiang province, including 24 males and 30 females. EGFR and K-ras gene mutations was examined by PCR and bi-direction sequencing. Results EGFR mutation occurred in 37.0% (20/54) of lung adenocarcinoma specimens, including mutations at exon 19 in 14 cases and exon 21 in 6 cases. K-ras mutation was detected at exon 19 and 21 in 1 case (2.0%). EGFR gene mutation was not correlated with sex, age, tumor location, lymph node metastasis and tumor differentiation. Conclusion There is no correlation of EGRF gene mutation with sex, age and other clinicopathological features in lung adenocarcinoma patients in Zhejiang province, and K-ras mutation is rare.
