摘  要:目的制备mB7.1-GPI膜表面修饰的结肠癌细胞疫苗,并探讨其抗肿瘤作用。方法将mB7.1-GPI融合蛋白锚定于小鼠结肠癌细胞膜上,制备结肠癌细胞疫苗,并采用流式细胞术检测该蛋白的细胞膜锚定效果;建立C57BL/6小鼠结肠癌模型,检测mB7.1-GPI融合蛋白肿瘤疫苗对荷瘤小鼠的免疫治疗作用。结果 mB7.1-GPI融合蛋白可锚定在小鼠结肠癌细胞膜上,并有效刺激小鼠脾细胞增殖和分泌IL-2和IFN-γ,mB7.1-GPI膜表面修饰的结肠癌细胞疫苗能够显著抑制荷瘤小鼠肿瘤的生长,并延长其生存期。结论 mB7.1-GPI融合蛋白制备的结肠癌细胞疫苗具有较强的抗肿瘤作用,可能成为一种有效的、用于结肠癌的治疗和预防的新型疫苗。Objective To prepare the mB7.1-GPI-surface-modified CMT93 colon cancer cell vaccine and investigate its anti-tumor effects. Methods mB7.1-GPI fusion protein was anchored on the membrane of colon cancer ceils to prepare the mB7.1-GPI-anchored colon cancer cell vaccine, and the anchoring effect of this protein was detected through flow cytometry. C57BL/6 mice models of colon cancer cell were established to observe the immunotherapeutic effects of mB7.1-GPI-anchored colon cancer cell vaccine on mice bearing tumor. Results mB7.1-GPI fusion protein was stably anchored on the membrane of CMT93 colon cancer cells and effectively induced the proliferation of spleen cells as well as the secretion of IL-2 and IFN-γ. The mB7.1-GPI-surface-modified CMT93 colon cancer cell vaccine was capable of significantly inhibiting the growth of tumors in mice bearing tumor and prolonging their survival durations. Conclusion The tumor vaccine prepared by mB7.1-GPI fusion protein has effective anti-tumor effects. It may be used as a new type of cell vaccine in the prevention and treatment of colon cancer.
