摘  要:目的 探讨外周血淋巴细胞 (PBL)中金属硫蛋白 (MT)基因表达作为镉接触生物标志物的可行性。方法 选择职业接触镉的人群 ,进行接触评定和效应评定 ,分离淋巴细胞进行体外培养 ,测定MT基因表达水平。结果 镉接触工人PBL的基础、诱导的MT表达水平分别为 1.5 7±0 .5 7、3.6 2± 1.0 2 ,明显高于对照组 (1.2 4± 0 .45、2 .6 8± 0 .78) ,差异有显著性 (P <0 .0 5 )。随尿镉、血镉和镉摄入量水平上升 ,镉接触工人PBL的MT基础表达和诱导表达都增高。MT基础和诱导表达与血镉水平显著相关 (偏相关系数分别为 0 .493和 0 .5 91) ;同时与尿镉也显著相关 (偏相关系数分别为 0 .314和 0 .334)。血镉水平与MT基础表达和诱导表达存在剂量 -效应关系。Objective To investigate the feasibility of metallothionein(MT) gene expression level in peripheral blood lymphocyte(PBL) as a biomarker of cadmium exposure.Methods MT gene expression level in PBL of workers exposed to cadmium was examined using dRT PCR technique,and exposure assessment and effect assessment were conducted in exposed workers.Results It was shown that basal and induced expression levels in PBL of exposed workers were 1.57± 0.57 and 3.62±1.02 respectively,higher than those of control group(1.24±0.45,2.68±0.78, P <0.05).There was a dose effect relationship between MT gene expression level and blood cadmium level( r =0.493, r =0.591).The former was also correlated with urine Cd. Conclusion The results suggested the feasibility of MT gene expression in PBL as a biomarker of cadmium exposure.
