摘 要:目的建立大鼠颈上交感神经节细胞的急性方分离法并鉴定其电生理学特性。方法sD大鼠,雌雄不拘,5~12d龄,断头后,急性分离大鼠的颈上交感神经节,经酶解消化法获得颈上交感神经节细胞。采用全细胞膜片钳技术,于钳制电位-60mv下,加入100,umol/L乙酰胆碱,记录乙酰胆碱受体电流。采用碳纤维电极技术,于钳制电位760mV下,加入65mmol/LKCl,检测儿茶酚胺类物质分泌活动。结果分离出表面光滑、遮光性较好的颈上交感神经节细胞,并记录到乙酰胆碱受体电流和儿茶酚胺类物质的分泌。结论急性分离大鼠颈上交感神经节细胞的方法可获得适于膜片钳实验的细胞。Objective To establish a method for acute isolation of rat superior cervical ganglion (SCG) cells and identify the electrophysiological properties. Methods Sprague-Dawley rats of both sexes, aged 5-12 days, were decapitated. The SCGs were removed quickly, and the single SCG cell was enzymatically isolated from the SCGs. When the holding potential was - 60 mY, 100μmol/L acetylcholine was applied and the nicotinic ace-tylcholine receptor currents were recorded by whole-cell patch-clamp technique. When the holding potential was 760 mV, 65 mmol/L KC1 was applied and quantal release of catecholamines was detected by using carbon fiber electrodes. Results SCG cells with normal electrophysiological properties were isolated. Typical nicotinic acetyl-choline receptor currents and quantal release of catecholamines were recorded successfully. Conclusion The ceils suitable for patch-clamp experiments can be obtained by using the method for acute isolation of rat SCG cells.
