摘  要:目的:观察通经补肾中药方对铝诱导的阿尔茨海默病(AD)模型大鼠氧化应激(OS)的影响。方法:选择健康3月龄SD大鼠84只,随机分为对照组(C组)、模型组(按剂量从小到大分为M1、M2、M3组)、中药组(按剂量从小到大分为N1、N2、N3组),每组12只。模型组在常规饲料中添加不同剂量AlCl.36H2O喂饲大鼠,持续染毒3个月,制作AD动物模型。用电迷宫测试大鼠学习记忆能力,模型组大鼠对电刺激的逃避潜伏期明显延长,逃避正确率显著降低,证明AD模型制备成功。中药组大鼠喂饲高剂量铝饲料,并以不同剂量通经补肾中药方灌胃。上述实验结束后,检测大鼠脑组织及血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-Px)活性和丙二醛(MDA)水平。结果:与C组比较,M2、M3组大鼠脑组织及血清中SOD、GSH-Px的活性均明显降低(P<0.01),MDA含量显著增高(P<0.01)。M1组与C组,M2组与M3组各指标比较差异均无统计学意义(P>0.05)。与M3组比较,N2、N3组大鼠SOD、GSH-Px的活性明显增高(P<0.01),MDA含量明显降低(P<0.01)。N1组与M3组,N2组与N3组各指标比较差异均无统计学意义(P>0.05)。结论:铝的过多摄入使大鼠处于强氧化应激状态,通经补肾中药方可显著提高模型大鼠抗氧化能力,从而对中枢神经系统有保护作用。Objective: To observe the effect of Tongjing Bushen recipe on the oxidative stress of Alzheimer's disease (AD) model rats induced by A1. Methods: Eighty-four healthy SD rats, 3-month old, were randomly divided into control group (group C), model group (including low-dose A1 group-M1, middle-dose A1 group-M2 and high-dose A1 group-M3) and Chinese-medicine-recipe group (including low-dose group-N1, middle-dose group-N2 and high-dose group-N3). There were 12 rats in each group. Rats in model group were fed with conventional feed mixed with A1C136H20 for three months to establish the AD model. The successful AD animal model was proved by detecting the learning and memory ability with Y-electric maze. The escaping latent period was obviously prolonged, and the correct rate of escape was significantly decreased in group M compared with that of control group. Rats in Chinese medicine group were fed with high-dose A1 and Tongjing Busben recipe. The levels of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and malondialdehyde (MDA) in serum and brain tissues were detected and compared between groups. Results: Compared with group C, the activities of SOD and GSH-PX in brain tissues and serum were strikingly decreased, the content of MDA was strikingly increased,in group M2 and group M3 (P 〈 0.01). There were no significant differences in each index between group M1 and group C,and between group M2 and group M3 (P 〉 0.05). The activities of SOD and GSH-PX were significantly increased, and the content of MDA significantly decreased in group N2 and group N3 compared with those of group M3 (P 〈 0.01). There were no significant differences in each index between group N1 and group M3, and between group N2 and group N3 (P 〉 0.05). Conclusion: The excessive intake of A1 caused the state of intense oxidative stress in rats. Tongjing Bushen recipe strikingly increased the antioxidant ability and protected the Central nervous system in rats.
