摘 要:采用水提醇沉法制备仙人掌多糖(简称OPS),并分析三氯乙酸脱杂蛋白的方法对多糖抑菌作用的干扰;采用管碟法和混合涂布法研究OPS及仙人掌粉对食品中E.col(i大肠肝菌)、S.aureus(金黄色葡萄球菌)、S.cerevisiae(酿酒酵母)和P.frequentans(青霉)等常见污染菌的抑菌效果。结果发现,三氯乙酸会造成OPS的假阳性抑菌反应,利用乙二醇可以将OPS中的三氯乙酸去除;与E.coli和P.frequentans相比较,OPS及仙人掌粉对S.aureus和S.cerevisiae的抑菌效果明显;混合涂布法较管碟法更适宜检测OPS等水溶液具有一定黏稠度的抑菌剂的抑菌效果。The conventional method of water extraction and ethanol precipitation was applied to prepared polysaccharide from opuntia (OPS) powder. The interference of trichloroacetic acid to antibacterial activity of OPS which was used to eliminate the foreign protein was discussed. The inhibition ability of OPS and opuntia powder to some common contaminated microbes was studied by means of tube disc method and mixture spread plate method. The results showed that trichloroacetic acid was the mainly factor to false positive antibacterial reaction of OPS. It could be removed away OPS by using glycol solvent. The antibacterial activity of OPS and opuntia powder to S.aureus and S.cerevisiae was superior to E.coli and P. frequentans. With respect to viscous bacteriostatic agent, the mixture spread plate method was preferable to the tube disc method.
