摘 要:目的比较不同背景的前列腺癌小鼠(transgenic adenocarcinoma mouse prostate,TRAMP)模型的病理差异并初步探讨其意义。方法比较24周龄TRAMP和TRAMP×FVB小鼠的泌尿生殖系统的相对质量;利用HE染色比较24周的TRAMP和TRAMP×FVB小鼠的肿瘤病理进程的差异,采用免疫组织化学方法检测前列腺组织细胞增殖核抗原(proliferating cell nuclear antigen,PCNA)、血管性假血友病因子(von willebrand factor,vWF)和大T抗原(large tumor antigen,Tag)的表达。结果 TRAMP×FVB小鼠前侧前列腺(anterior prostate,AP)的病理等级与TRAMP小鼠比较差异无统计学意义,但背部前列腺(dorsalprostate,DP)、腹部前列腺(ventral prostate,VP)、旁侧前列腺(lateral prostate,LP)的病理等级要明显低于TRAMP小鼠;TRAMP×FVB小鼠PCNA和vWF的表达高于TRAMP小鼠;两种背景小鼠的Tag基因表达及转移情况无明显差异。结论 TRAMP×FVB小鼠前列腺癌的病理进程快于TRAMP小鼠,其病理进程的加快与微血管密度增加及处于增殖期肿瘤细胞数目的增加密切相关。Objective To compare the pathological difference between transgenic adenocarcinoma mouse prostate (TRAMP) and TRAMP x FVB mice. Methods The relative quality of genitourina17 system was observed at 24 weeks in two kinds of mice. Tumor progression was determined by histopathological examination at 24 weeks. The expression of proliferating cell nuclear antigen ( PCNA), von willebrand factor (vWF) and large tumor anligen (Tag) was decided by immunohistoehemical staining. Results Compared with TRAMP mice, the pathological grade of dorsal prostate ( DP), ventral prostate ( VP), lateral prostate (LP) is more severe in TRAMP~ FVB mice, as well as the expression of PCNA and vWF was increased. Both two kinds of mice showed no difference in T antigen expression anti tumor metastasis. Conclusion Tumor progression was more prompt in TRAMP x FVB nfice than TRAMP mice, which may be related to increasing nficrovascular density and tumor cells proliferation.
