摘　　要：目的观察顺铂(DDP)在体外对人皮肤鳞癌细胞系A431和皮肤恶性黑素瘤A375细胞的毒性作用,为临床化疗提供参考。方法将A431和A375细胞株分别在含有不同浓度DDP培养基中孵育,采用四甲基偶氮唑盐法(MTT)测定细胞的抑制率。结果 DDP对A431及A375细胞的抑制率随着药物浓度增加而增加;高浓度DDP对A431抑制率更高(P<0.05)。结论 DDP对A431及A375具有细胞毒性,在实验条件下A431对高浓度DDP更敏感。Objective To study the cytotoxicity of cisplatin（DDP） in skin cancer cell lines in vitro and provide references for drug therapy in clinical practice.Methods A431 cell lines or A375 cell lines were incubated in vitro with different concentrations of DDP,and their inhibitive rates were tested with MTT assay.Results Killing effects of DDP on A431 cell lines or A375 cell lines were enhanced as the concentrations of DDP had been increased.High-dose DDP had a more significant killing effect on A431 cell lines（P〈0.05）.Conclusion DDP is cytotoxic in A431 cell lines and A375 cell lines.A431 cell lines are more sensitive to DDP compared with A375 cell lines in the experimental condition.
