脂肪源性干细胞穴位注射对大鼠脑缺血再灌注后胰岛素样生长因子1及其受体表达的影响

摘　　要：目的探讨脂肪源性干细胞(ADSC)穴位注射对大鼠缺血/再灌注损伤后胰岛素样生长因子1(IGF-1)及其受体(IGF-1R)表达的影响。方法线栓法制作大鼠大脑中动脉缺血(MCAO)2 h再灌注模型,大鼠随机分为假手术组、模型组、穴位注射ADSC组(治疗组)、穴位注射0.9%氯化钠注射溶液组(对照组)。治疗组取在大椎、双侧内关穴位注射PKH-26标记ADSC细胞悬液。缺血72 h后NSS法检测神经功能恢复,采用实时荧光定量PCR及免疫组织化学方法检测IGF-1、IGF-1R蛋白和mRNA的表达。结果治疗组可见PKH26标记的ADSC在海马区有表达。与模型组和对照组比较,穴位注射ADSC组NSS评分显著降低(P<0.05),海马区IGF-1、IGF-1R蛋白和mRNA表达显著增加(P<0.05)。结论穴位移植脂肪源性干细胞对大鼠脑缺血再灌注有神经保护作用,其作用机制可能与促进海马区IGF-1和IGF-1R表达有关。Objective To investigate the effect of acupoint injection with adipose-derived stem cell on insulin-like growth factor 1（IGF-1） and receptor（IGF-1R） after cerebral ischemia-reperfusion in rats.Methods The rat model of middle cerebral artery occlusion was established in model.A total of 48 healthy SD rats were randomly divided into sham operation group,model group,ADSC acupoint injection group（treatment group） and saline acupoint injection（control group）.ADSC suspensions were marked with PKH-26 were infused into acupoints of ＂Dazhui＂and＂Neiguan＂ in the treatment group.72 h after reperfusion,neuological function score were observed by NSS.The expressions of IGF-1 and IGF-1R were observed by using immunohistochemical and quantitative real-time RT-PCR.Results PKH-26 labeled ADSC were observed in hippocampus region of the treatment group.Compared with model and control group,the NSS and the expression of IGF-1,IGF-1R of the treatment group were significantly increased（P0.05）.Conclusion ADSC acupoint injection could improve the neurological function in rats after cerebral ischemia-reperfusion,which may be related to up-regulating IGF-1 and IGF-1R in hippocampus region.
