摘 要:目的观察Aβ25~35海马注射对大鼠海马硫化氢(hydrogensulfide,H2S)/胱硫醚β-合酶(cystathionine-β-syn-thase,CBS)体系产生的影响。方法通过双侧海马内注射Aβ25~35制作大鼠AD模型,对照组双侧海马内注射生理盐水。采用全自动酶标仪测定各组大鼠海马中H2S含量、CBS活性。结果 AD组与生理盐水组比较,海马H2S含量均明显降低(P<0.01),CBS活性显著增高(P<0.05)。结论 Aβ25~35海马注射可导致大鼠海马H2S水平下降,CBS表达增强,内源性H2S与CBS之间可能存在负反馈调节作用。Objective To investigate the effect of microinjection of Aβ25 ~35 on hydrogen sulfide ( H2S) /cystathionine-β-syn-thase ( CBS) system in the hippocampus of rats. Methods A model of AD was induced by microinjection of Aβ25 ~ 35 into thehippocampus in SD rats. And the NS group induced by microinjection of normal saline into the hippocampus. The H2S levels,andCBS activities in hippocampus in three groups were determined using a Universal Microplate Reader. Results Compared with NSgroup,H2S content in hippocampus was significantly decreased ( P 0. 01) while the expression of CBS activities was up-regulated( P 0. 05) in AD group. Conclusion microinjection of Aβ25 ~35 into the SD rats hippocampus can decrease H2 S level and up-regulate the expression of CBS activity in thehippocampus of rats. There maybe negative feedback effect between H2 S and CBS in ratshippocampus.
