摘　　要：目的探讨重楼皂苷I（PolyphyllinI，P-I）对人卵巢癌细胞株SKOV3的体外生物学效应。方法以不同浓度的P-I对SKOV3细胞作用不同时间后，分别应用CCK-8比色法和流式细胞术检测细胞体外增殖和周期改变，并用Hoechst 33258荧光染色法和AnnexinV-PI双染法检测细胞凋亡变化。结果在一定浓度范围，P-I抑制SKOV3细胞体外生长增殖呈明显的量-效关系，与对照组比较差异具有明显统计学意义（P〈0．01）；P-I在SKOV3细胞分裂周期的G0／G1期具有高度作用敏感性；P-I明显诱导SKOV3细胞发生凋亡。结论P-I可能是通过干扰人卵巢癌细胞株DNA合成，抑制其生长增殖，并诱导凋亡，从而起到抗卵巢癌的作用。Objective To define the biological effects of human ovarian carcinoma SKOV3 cells line （SKOV3 cells）in vitro after treatment with polyphyllins I （P - I）. Methods The proliferation - inhibiting effect and cycle of SKOV3 cells which were treated with different concentrations of P - I for different times in vitro were detected by CCK -8 colorimetric and flow cytometry respectively, and the cell apoptosis was examined by Hoechst 33258 fluorescent staining and Annexin - IP double staining. Results In a certain range of P - I concentration, the proliferation and growth of SKOV3 cells in vitro were inhibited which showed a clearly dose - effect relation- ship, and the difference existed statistically significance when compared with the control group （P 〈 0.01 ） ;P -I had high sensitivity on G0/GI phase of SKOV3 cells cycle ; and the results showed that P - I induced apoptosis in SKOV3 cells. Conclusion P - I may interfere DNA synthesis, inhibit proliferation and induce apoptosis in hu- man ovarian cancer ceils, thus playing a role in anti -ovarian cancer.
