Survivin基因在不同胃黏膜中的表达的研究

摘　　要：目的:研究胃癌(gastric cancer,GC)及不同胃黏膜组织中生存素(Survivin)基因的表达关系,为临床应用以生存素为靶点的胃癌防治措施提供理论依据。方法:采用免疫组织化学染色(SP法)检测不同胃黏膜组织中Survivin。30例浅表性胃炎(chronic superficial gastritis,CSG);31例萎缩性胃炎(chronic Atrophic gastritis,CAG),15例异型增生(Dysplasia,Dys)。胃癌组织32例。结果:从CSG→CAG→Dys→GC过程中,Survivin表达均呈递增趋势。CSG灰度值(57.64±4.44)、CAG灰度值(55.05±6.97)、Dys灰度值(48.59±4.16);GC灰度值(32.25±4.13)。Survivin表达在CSG、CAG、Dys、GC之间表达有统计学意义(P<0.05)。胃癌组织Survivin的表达与年龄,性别,肿瘤大小无关;在临床分期和组织分化程度方面Survivin阳性表达率明显不同,具有统计学意义(P<0.05)。结论:Survivin基因在胃癌的发生发展过程中可能起重要作用。Objective：To investigate the expression of Survivin in various gastric mucosa and analyzed its relationship,in order to provide a novel target to treat GC.Methods：The expression of Survivin in various gastric mucosa was detected by immunohistochemistry methods（sp-methods）in 30 human chronic superficial gastritis（CSG）,31 human chronic atrophic gastritis（CAG）,15 dysplasia（Dys）and 32 human GC tissues.Result：Expression of telomerase showed a gradually increasing tendency from CSG,CAG,Dys to GC.The values of gray scale in CSG,CAG,Dys and GC groups were 57.64±4.44,55.05±6.97,48.59±4.16 and 32.25±4.13 respectively.The expression level of Survivin showed significant difference among CSG,CAG,Dys and GC group（P〈0.05）.Survivin protein was correlated with clinical staging and differentiation status（P〈0.05）,but had no obvious relationship with age,sex,size of tumor.Conclusion：Survivin was overexpressed in gastric cancer.Survivin gene may play an important role in the course of tumorigenesis and progression in gastric carcinoma.
