PTEN、survivin基因表达与胃癌化疗药物敏感性的相关性研究

摘　　要：目的研究原代培养胃癌细胞对化疗药物的敏感性,并进一步探讨其敏感性与胃癌组织中PTEN(phos-phatase and tensin homolog deleted on chromosome ten)及survivin基因表达的关系,为胃癌的临床化疗提供个体化用药的实验资料和依据。方法取手术切除的新鲜胃癌组织标本,用MTT法检测经羟基喜树碱(HCPT)、顺铂(DDP)、阿霉素(ADM)、5-氟尿嘧啶(5-FU)、丝裂霉素(MMC)药物作用后肿瘤细胞活力及代谢活性的变化;同时取胃癌组织制成石蜡切片,用免疫组化检测PTEN、survivin蛋白的表达。结果 PTEN阳性表达与五种化疗药物的敏感性无关(P<0.05)。survivin阳性表达与5-FU及ADM耐药有关(P<0.05);survivin阳性表达与PTEN蛋白阳性表达呈负相关关系(rs=-0.668,P=0.000);结论 1、胃癌组织中survivin阳性表达者与5-FU及ADM耐药有关,提示survivin可作为胃癌原发性耐药标志之一。2、胃癌组织中PTEN的表达与survivin的表达呈负相关关系,提示两者在胃癌的发展中可能具有协同作用,两基因除各自直接作用外还可通过负性调节对方基因的表达而间接发挥功能。Objective： To evaluate the chemosensitivity of primary culturing gastric tumor cells by using MTT and the protein expressions of PTEN and survivin in gastric cancer tissues,and to explore the relationship between PTEN or survivin expression and chemosensitivity of gastric cancer.It is also to determine whether these results can be used to help therapy the gastric carcinoma.Methods： Inhibitory rate of cells was detected by MTT assay.Expression of PTEN and survivin in gastric cancer tissues were detected by immunohistochemistry（IHC）.Results： Expression of PTEN protein was significantly negatively related to that of survivin（rs=-0.668,P=0.000）.The positively survivin stained cells showed resistance to 5-FU and ADM.Conclusion： 1.The survivin positive cells showed resistance to 5-FU and ADM.Our results suggest that survivin positive expression may contribute to primary drug-resistance of tumor.2.Expression of PTEN protein is significantly negatively related to that of survivin.This result proves that PTEN plays the function of restraining cancerization by regulating nagetively the expression of survivin besides by its own action.
