摘 要:利用抗肿瘤相关抗原的抗体对肿瘤进行导向定位、治疗已引起医学界的普遍关注。核素标记的抗肝癌单抗的研究始于近年,国内尚无完整报道。本文用131I标记的抗人肝癌单抗在裸鼠人肝癌模型上进行肿瘤定位,并分析了影响定位的因素及进一步改进的途径。Localization of human hepatocellular carcinoma (HCC) xenografts in nude mice hasbeen studied by using ^(131)I-labeled monoclonal antibody Hepama-Ⅰ against HCC cellline BEL-7402. Gamma camera picture showed that there was obviously specific uptakeby the tumor cells at day 4 post-injection of ^(131)I labeled Hepama-1 (^(131)I-Hepama-Ⅰ).The radioactivity in tumor center was 9.8 times higher than that in the peripheraltissues and was 5.3 times higher than that in tumor center in the control group (^(131)I-IgG) In ^(131)I-Hepama-Ⅰ group, the tumor/liver ratio increased from 1.3 to 3.03, anduptake rate of the tumor cells increased continuously from 11.3%/g to 21.5%/g overday 1 to day 7. But the tumor/liver ratio in ^(131)I-IgG control group was less than 1, theradiative uptake rate of tumor cells is not higher than 11.95%/g at day 4 and decreasedpromptly to 7.1%/g at day 7.
