摘 要:人肿瘤的发生与某些原癌基因的活化有密切关系,ras族和myc族基因是人类肿瘤常见的癌基因。许多人曾报道在人的肺癌组织或细胞株中有Ki-ras或/和C-myc基因的放大、突变活化或异常表达。癌基因是通过其产物而起生物学作用的,为了探索癌基因产物在癌变过程中的作用及其应用价值,首先必须得到大量的基因产物,我室曾构建了V-Ki-ras和V-myc的表达质粒。本文主要报道抗V-Ki-ras、V-myc基因产物多克隆抗体的制备及应用多克隆抗体以免疫印迹法检测女性肺癌组织中Ki-ras和C-myc的产物。Immunoblot analysis was carried out to test for Ki-ras and c-myc products of the35 samples of lung cancer and cancer-surrounding tissues from 18 female patients. Theresults suggested that Ki-ras encodes p21 while c-myc expresses p58 (also indicated byblock experiment). The cancer tissue samples from 13 patients (72%) were observed tobe p21-positive while the same number of patients were p58-positive. Thus, the rates ofp21 and p58 occurrence (not their coincidence) among lung cancer patients appeared tobe similar. In the majority of patients tested, however, cancer and cancer-surroundingtissues showed p21-positive simultaneously. With p58-positive signals the case wasthe same. It is noteworthy that in the same cancer tissue the concomitant over expres-sion of p21 and p58 was identified in up to 61% of the samples suggesting that activa-tion of these two proto-oncogenes might occur concomitantly.
