压应力对Chang肝细胞株中肝再生因子表达的影响

摘  要:目的探讨压应力对Chang肝细胞株肝再生因子如核因子-κB(NF-κB)、信号转导子与转录激活因子3(STAT3)、原癌基因c-fos及c-jun等表达变化的影响。方法给予人Chang肝脏细胞株1000μ和2000μ压应力的作用,诱导肝细胞再生,分别在30 min、1h、2h、3h、4h、6h应用Western blot分析检测NF-κB P65、p-STAT3,RT-PCR方法检测c-fos和c-jun的表达。结果压应力作用后在人Chang肝细胞株中,Western blot分析示NF-κB P65、p-STAT3的表达迅速升高,压应力作用量大小对NF-κB P65仅在2h时点差异有统计学意义(P=0.046),对p-STAT3的表达仅在1h时点表达差异有统计学意义(P=0.039),二者表达高峰分别在1h、30min;RT-PCR示不同大小的压应力对c-fos表达无影响,且各时间点表达无差异,而对c-jun的表达在30min(P=0.026)、1h(P=0.031)、2h(P=0.033)差异有统计学意义。结论压应力可能在肝细胞再生的起始阶段发挥着重要的作用。Objective To investigate the effect of compressive stress on the factors for liver regeneration including NF-κB, signal transducers and activators of transcription 3 (STAT3), c-fos and c-jun in hepatocytes Chang cell line. Methods Human hepatocytes Chang cell line were subjected to compressive stress at 1000 μstain or 2000 μstain, expression of NF-κB P65, p STAT3, c-los and c-jun were detected by Western blot or RT-PCR at 30 min, 1 h, 2 h, 3 h, 4 h, 6 h after application of compressive stress to indicate the priming of hepatocytes proliferation. Results Enhanced expressions of NF-κB P65 and p-STAT3 were observed in hepatocytes under compressive stress indicated by Western blot, the magnitude of compressive stress loaded significantly affected the level of expression of NF-κB P65 at 2 h (P=0. 046) and p STAT3 at 1 h (P=0. 039), the peak of expression of p- STAT3 was at 30 minutes after stress-loading while NF-κB P65 was at 1 hour; RT PCR showed that expression of c-fos was not significantly different between 1000 μstain and 2000 μstain at each time point, and expression of c-jun was significantly different at 30 minutes (P=0.026), 1 h (P=0.031), 2 h (P=0.033) after compressive stress loading. Conclusion These results indicate that compressive stress may play an important role in initiating the process of liver regeneration.
