人胃癌HGC-27细胞及其肿瘤球细胞裸小鼠皮下移植的研究

摘　　要：目的:研究人胃癌HGC-27细胞及其肿瘤球细胞在裸小鼠皮下产生肿瘤的能力。方法:用DMEM培养液(SSM)培养HGC-27细胞。用含有生长因子的DMEM/F12培养液(SFM)在低吸附6孔培养板中诱导肿瘤球的生长。分别将溶于200μL PBS的1×102、1×103、1×104、1×105、1×106和1×107肿瘤细胞或肿瘤球细胞注射到裸小鼠皮下。常规方法制作组织切片。结果:在SSM培养液中HGC-27细胞呈贴壁、伸展状态生长。在SFM及低吸附的培养条件下观察到了肿瘤球的形成,呈悬浮状态。注射了1×106、1×107肿瘤细胞的裸小鼠产生了移植瘤,注射了1×104、1×105、1×106和1×107肿瘤球细胞的裸小鼠产生了移植瘤。移植瘤组织癌细胞间连接疏松,瘤细胞分化程度低。结论:HGC-27肿瘤球细胞比非肿瘤球HGC-27细胞具有更强的致瘤能力。OBJECTIVE： To study the tumorigeness of HGC-27 cells and tumor sphere cells in nude mice. METH- ODS： HGC-27 cells were cultured in SSM. Tumor spheres were induced by SFM in 6 well. cell culture cluster. 1×10^2、1×10^3、1×10^4、1×10^5、1×10^6 HGC 27 cells and tumor sphere cells were transferred into nude mice. The histo- logical section was made with routine method. RESULST： The HGC-27 cells were adherent and spreading in SSM. The tumor spheres were seen inSFM. 1×10^6,1×10^7 HGC-27 cell formed tumor in nude mice and 1×10^4,1×10^5,1×10^6, 1×10^7 tumor sphere cell formed tumor were observed in nude mice. The conjunction of cancer cells was loose in histologi- cal section. CONCLUSION： Tumorigeness of tumor sphere cell is higher than that of HGC-27 cell.
