摘  要:比较陕西、山西、云南、四川等地野生猪苓中麦角甾醇含量。野生猪苓样品经甲醇超声提取,用HPLC进行测定。采用Phenomenex C18(250mm×4.60mm,5μm)色谱柱,流动相为甲醇,流速为1.0mL.min-1,检测波长为283nm,柱温30℃。麦角甾醇进样量在0.26—2.64μg范围内线性关系良好(r=0.9999)。平均加标回收率(n=3)分别为102.18%、100.09%、97.84%;RSD分别为4.94%、4.48%、4.24%。测定的18个野生猪苓样品中除个别样地麦角甾醇含量低于2010版《中国药典》规定麦角甾醇最低含量0.05%外,其他均符合药典规定。不同产地猪苓中麦角甾醇含量存在显著性差异(P<0.05),结合野生猪苓形态学记录以及生长环境分析,推测猪苓中麦角甾醇的含量高低可能与猪苓有效生长年龄和密环菌有关,其具体作用机制还有待进一步研究。The contents of ergosterol in wild Polyporus umbellatus(Pers.) Fries from Shaanxi,Shanxi,Yunnan,Sichuan were compared.The Polyporus umbellatus(Pers.) Fries were extracted by ultrasound with methanol,and was determined by HPLC.The compound was separated by Phenomenex C18(250mm×4.60mm,5μm) column with mobile phase of methanol at a flow rate of 1.0mL·min-1.The detection wavelength was 283nm and column temperature was 30℃.The sample size of ergosterol had the good linear relationship in the range of 0.26—2.64μg(r=0.9999).The average recoveries were respectively 102.18%,100.09% and 97.84%(n=3),and RSDs were 4.94%,4.48% and 4.24%,respectively.Except for one exceptional ergosterol was lower than the lowest ergosterol content of 0.05% provided by the 2010 version of "Chinese Pharmacopoeia",the determination results of all the others in 18 different wild Polyporus umbellatus(Pers.) Fries were according with the Pharmacopoeia.There was the significant difference in Polyporus umbellatus(Pers.) Fries from the different producing areas(P〈0.05).Combination of morphological records and growing environment analysis of wild Polyporus umbellatus(Pers.) Fries,the level of ergosterol content in Polyporus umbellatus(Pers.) Fries may be related to the effective growth age of Polyporus umbellatus(Pers.) Fries and the dense of armillaria mellea.The specific mechanism remains to be further studied.
