摘  要:建立了一种利用阴离子交换色谱柱分离,抑制型电导检测器同时测定乳酸、丙酸、丙酮酸、苹果酸、磷酸二羟丙酮、柠檬酸、异柠檬酸7种有机酸和单磷酸核苷酸(AMP)、双磷酸核苷酸(ADP)、三磷酸核苷酸(ATP)3种核苷酸的方法。采用IonPacAS11-HC阴离子交换柱,KOH梯度洗脱,流速1.00mL/min,柱温30℃,进样体积25μL。方法的相对标准偏差为0.39%~4.69%,线性相关系数为0.9906~0.9996,加标回收率为80.71%~94.02%,方法可用于嗜热厌氧菌胞外培养液中有机酸和核苷酸的同时测定。This paper introduced a new method for rapid determination of several kinds of phosphates in scallop. Before analysis, the samples were ultrasonically extracted and purified using solid-phase extraction column.The resulting supernatants were analyzed by ion chromatography with a high-capacity anion exchange column and suppressed conductivity detection. The effects of experimental conditions on the testing results for a variety of phosphates were discussed. The experimental results shows that the limit of detection was 40 mg/kg for sodium phosphate and 10 mg/kg for sodium pyrophosphate, sodium tripolyphosphate and sodium trimetaphosphate. The recovery was above 74.8% and the RSD was less than 10.7%. The results for analyzing the real samples are satisfied. This method has the advantages of simplicity, short testing time, good selectivity, high sensitivity, no pollution effect to environment, and simplicity of the operational procedure, etc.
