摘　　要：目的研究DPC4基因转染对结肠癌SW480细胞的影响。方法建了表达DPC4基因的逆转录病毒pLXSN载体,转染SW480细胞和未转染的SW480细胞为对照。Western鉴定DPC4蛋白的表达,MTT法检测细胞增殖率。结果发现SW480/DPC4细胞较SW480/pLXSN、SW480/-细胞,生长明显减缓,经统计有显著差异(P<0.05)。培养第7天的时候,抑制率达50%左右。结论 DPC4基因具有抑制结肠癌细胞生长的作用,为结肠癌的侯选肿瘤抑制基因。Objective To investigate the Effects of DPC4 gene on the SW480 cells. Methods The retroviral vector pLXSN containing DPC4 which could be used to transfect the colorectal carcinoma cells SW480 was constructed,with the empty vector pLXSN as a positive control. Results Compared with those of the controls,the cell in vitro prolifera- tion was assessed by MTT assay. Conclusion DPC4 could mediate tumor suppression through suppression of angiogen- esis.
