摘  要:目的观察整合素β3在胶质瘤肿瘤干细胞(GSCs)的肿瘤血管生成中的作用。方法 分离培养GSCI和GSC2,制备人间质干细胞(hMSCs)条件培养基(hMSC—CM),分别在神经干细胞(NSC)培养基和hMSC—CM中培养,观察管样结构形成结果;通过Westernblot法检测NSC和hMSC—CM培养条件下整合素的表达;通过脂质体转染小干扰RNA(siRNA)对GSC1的整合素β3进行RNA干扰,检测RNA干扰后GSCl的管样结构形成。结果GSCI在hMSC—CM培养下可以观察到类似管佯结卡勾彤成,Westernblot法检测结果显示GSCI存hMSC—CM培养7d时整合素β3表达明显高,iNSC培养,siRNA干扰可以下调GSC1的整合素β3表达,影响了GSC1在hMSC—CM中形成管样结构。结论整合素β3在GSC1体外血管生成中起重要作用。Objective To investigate the role of integrin expression in the tube formation of glioma stem cells in vitro. Methods Glioma stem yells (GSCs) , GSC1 and GSC2 were isolated and mdtured from freshly surgically resected specimens of patients with glioblastoma. GSCs were maintained in neural stem cell (NSC) media supplemented with DMEM/F12 with EGF (20 μg/L), basic fibroblast growth factor2 (bFGF2) (20 μg/L) and 2% B27, 1% N2. Human mesenehymal stem cell conditioned media (hMSC-CM) were made. GSCs were cultured in NSC media and hMSC-CM separately to cheek the tube firmation. The integrins expression was dectected by using Western blotting. The small interfering RNA (siRNA) against the integrin β3 was transfected into the GSC1 to knock down the expression of integrin β3. The GSC1 with integrin β3 knock-down was seeded on the plate to check the tube tormation in vitro. Results GSC1 could form the tube formation in hMSC-CM after cuhure for 3 days. The integrin β3 expression of GSC1 in BMSC-CM was higher than that in NSC media. Knueking down the integrin β3 expression in GSC1 inhibited the tube formation in hMSC-CM. Conclusion lntegrins play important roles in glioma bMogy, including migration, invasion, and angiogenesis in vitro.
