摘  要:目的观察丙戊酸钠(VPA)对胰腺癌细胞NKG2D配体MICA和MICB表达的影响。方法1mmol/L的VPA孵育胰腺癌细胞24h,流式细胞术及实时定量聚合酶链反应(Real-timePCR)检测VPA处理前后胰腺癌细胞株PC-2及BxPC-3细胞表面NKG2D配体MICA和MICB的表达。结果胰腺癌细胞株PC-2及BxPC-3细胞表面MICA/B的阳性率分别为1.2%~1.5%、37.4%~42.5%;VPA处理后分别为3.2%~4.1%、62.4%~77.2%。PC-2及BxPC-3细胞MICAmRNA的表达水平为1.183±0.219、3.790±0.519,MICBmRNA的表达水平为1.020±0.025、3.267±0.183;经VPA处理后分别为3.173±0.456、16.700±0.534和2.543±0.385、14.730±0.370。结论VPA可以有效上调胰腺癌细胞株PC-2及BxPC-3细胞NKG2D配体MICA/B蛋白及mRNA的表达。Objective To study the effect of sodium valproate (VPA) on the expression of NKG2D ligands MICA and MICB in pancreatic cancer cells. Methods Pancreatic cancer cells were incu- bated with 1 mmol/L of VPA for 24 h, then, flow cytometry and real-time quantitative polymcrase chain reaction (Real-time PCR) were used to detect the expression of MICA/B in PC-2 and BxPC-3 cells of VPA-treated group and control group. Results The positive rate of MICAJB in PC-2 and BxPC-3 cells was 1, 2%-1.5% and 37.4%-42.5% ; and 3.2%-4. 1% and 62.4%-77.2% after VPA treatment. The MICA mRNA expression level in PC-2 and BxPC-3 cells was 1. 183 + 0. 219 and 3. 790 + 0. 519, and expression level of MICB mRNA was 1. 020+0. 025 and 3. 267 +0. 183. After VPA treatment, the MICA/B mRNA expression levels in PC-2 and BxPC-3 cells were 3. 173 +0. 456 and 16. 700 +0. 534, and 2. 543 +0. 385 and 14. 730 +0. 370 respectively. Conclusion VPA can effectively increase the expression of MICA/B in PC-2 and BxPC-3 cells.
