摘 要:目的通过对SFRP2基因在结直肠癌和癌旁组织中的超甲基化水平的测定,分析其与结直肠癌临床病理特征的关系。探讨SFRP2基因与结直肠癌浸润、转移等的关系。方法用实时定量聚合酶链反应技术检测分析30例结直肠癌患者癌组织、癌旁组织中SFRP2基因超甲基化水平,并统计学分析其与临床病理特征的关系。结果在癌组织和癌旁组织中均有SFRP2基因超甲基化发生;TNM分期中结直肠癌Ⅲ、Ⅳ期(7.24±1.13)组织中甲基化水平明显高于结直肠癌Ⅰ、Ⅱ期(5.92±0.97)组织中甲基化水平(P〈0.05);在低分化结直肠癌组织中(7.31±1.11)甲基化水平明显高于中高分化结直肠癌癌组织(6.23±1.03),差异有统计学意义(P〈0.05);与性别、年龄、肿块位置及大小差异无统计学意义(P〉0.05)。结论SFRP2基因超甲基化对结直肠癌恶性程度、浸润、转移和预后判定具有重要的意义。Objective To explore the differential level of hypermethylated SFRP2 gene in colorectal cancers and adjacent nontumorous tissues, to analyze the relation of SFRP2 gene promoter hypermethylation status and its clini- copathologic significance in colorectal carcinoma, and to study the relationship between the level of hyperrnethylated SFRP2 and the invasion and metastasis of coloreetal carcinoma. Methods Real-time quantitative PCR technique was performed to analyze the level of hypermethylated SFRP2 gene pronloter in colorectal cancers and adjacent non- tumorous tissues taken from 30 coloreetal cancer patients. The relation of the level of hypermethylated SFRP2 gene promoter and its clinicopathologic features of colorectal cancers was analyzed. Results SFRP2 gene promoter hypermethylalion occurred in both the tumur tissues and the adjacent tissues. The level of SFRP2 gene promoter hypermethylation was significantly higher in the patients with TNM Ⅲ and Ⅳ (7.24 ± 1.13)than in patients with TNM Ⅰ and Ⅱ (5.92 ± 0.97 ) ( P 〈 0.05 ). The level of SFRP2 gene promoter hypermethylation was significantly higher in the patients with lower differentiation (7.3 1± 1.11 ) than in patients with higher differentiation (6.23 ± 1.03 ) ( P 〈 0.05). No significant association was found between the level of SFRP2 gene promoter hypermethylation and the status of gender, age, tumor location and tumor size ( P 〉 0.05 ). Conclusions SFRP2 gene hypermethylation in tissues may be a useful ohjeetive parameter for the malignant level, invasion, metastasis, recurrence and prognosis of the colorectal cancer.
