摘 要:目的:观察通脑丸对慢性脑缺血性大鼠血清GSH-Px、MDA含量的影响。方法:采用双侧颈总动脉结扎法(2VO)造模,术后恢复性喂养3周,采用改进后的Bederson评分标准选取造模成功的36只大鼠,随机分成3组:模型组、通脑丸高剂量组和低剂量组,每组12只,空白组12只灌同等剂量的生理盐水,其余各组连续灌胃给药8周后,股动脉取血检测GSH-Px及MDA的含量。结果:模型组与空白组相比,血清GSH-Px活性明显下降(P<O.01);血清MDA含量明显升高(P<0.01),提示造模成功;通脑丸高、低剂量组与模型组相比,血清GSH-PX活性明显增强(P<0.01);血清MDA含量明显降低(P<0.01);而通脑丸高、低剂量组之间无统计学意义(P>0.05)。结论:通脑丸对慢性缺血脑组织具有保护作用,其机制可能通过提高GSH-PX活性,抑制缺血后MDA的升高而实现。Objective: To survey effects of TongNooWan on contents of GSH-Px and MDA in serum of rat with chronic cerebral ischemia. Method: Model was established through ligation of bilateral cephalic artery and was restored to feed for three weeks after surgery. Thirty-six rats were chosen according to reformed Bederson evaluation standards and randomly divided into three groups: model group, high, low dose groups of Tong Nao Wan, 12 rats each group. Twelve rats in blank group were administered with same doses of physiological saline while other groups were intragastrically administered with medicine for eight consecutive weeks. Contents of GSH-Px and MDA were detected through drawing blood from femoral artery. Result: Activity of serum GSH-Px obviously decreases (P〈0.01) and content of serum MDA rise significantly (P 〈0.01) when model group was compared with blank group, which suggested that the model was established successfully; when TongNooWan group (high and low dose groups) were compared with model group, activity of serum GSH-Px increased remarkably (P〈0.01); content of serum MDA reduced notably (P〈0.01); comparison between high and low dose groups of Tong Nao Wan showed no statistical meaning(P〉0.05). Conclusion: TongNaoWan can protect tissue of chronic cerebral ischemia, its mechanism might im- prove the activity of GSH-Px and inhibit the rising of MDA after ishchemia.
