摘  要:目的探讨基质金属蛋白酶-9(MMP-9)和金属蛋白酶组织抑制剂-1(TIMP-1)在甲状腺肿瘤组织中的表达。方法制备组织芯片,采用免疫组化和原位杂交技术检测56例甲状腺癌组织、癌旁组织和40例良性甲状腺病变组织中MMP-9和TIMP-1的表达情况。结果 MMP-9和TIMP-1蛋白阳性表达率在甲状腺癌组织中为71.4%和57.1%,MMP-9、TIMP-1 mRNA阳性表达率在甲状腺癌组织中为67.9%和62.5%,均明显高于癌旁和良性甲状腺病变组织中的阳性表达(P<0.05,P<0.01)。在甲状腺癌组织中,MMP-9、TIMP-1蛋白和MMP-9、TIMP-1mRNA的表达呈负相关性(R_(S1)=-0.309,R_(S2)=-0.264,均P<0.05)。结论 MMP-9和TIMP-1在组织中的检测有助于甲状腺癌的诊断,并可作为预后评估的重要参考。Objective To investigate the expression of matrix metalloproteinase-9(MMP-9) and inhibitor of melalloproteinase -1(TIMP-1) in the tissue of thyroid tumor.Methods The expressions of MMP-9 and TIMP-1 in 56 cases thyroid carcinoma tissues,56 cases adjacent tissues and 40 cases benign thyroid lesion tissues were detected by using immunohistochemistry and in situ hybridization,after preparation tissue microarray.Results The positive expression rates of MMP-9 and TIMP-1 protein were 71.4%and 57.1%respectively,and the positive expression rates of MMP-9 and TIMP-1 rnRNA were 67.9%and 62.5%respectively in the thyroid carcinoma tissues.They were all higher than the positive expression rates in adjacent tissues and benign thyroid lesion tissues(P 〈0.05,P 〈0.01 ).There were negative correlations between the expressions of MMP-9 and TIMP-1 protein(RS1 = - 0.309,P 〈 0.05 ) and between the expressions of MMP-9 and TIMP-1 mRNA (RS2 = - 0.264,P 〈 0.05) in the thyroid carcinoma tissues.Conclusion The detection of expressions of MMP-9 and TIMP-1 may not only contribute to differential diagnosis of thyroid carcinoma,but also help to predict the prognosis.
