摘  要:目的研究去甲二氢愈创木酸(NDGA)对骨肉瘤MG63细胞株黏附、迁移和侵袭特性的影响,并初步探讨其相关机制。方法不同浓度的NDGA处理MG63细胞,用CCK-8法、细胞划痕试验和Transwell小室模型分别测定其黏附力、迁移力和侵袭力,Western blot法检测MG63细胞基质金属酶(MMPs)表达量的变化。结果 NDGA明显抑制骨肉瘤MG63细胞株黏附、迁移和侵袭力(P<0.05);NDGA抑制MG63细胞的黏附、迁移和侵袭力呈剂量依赖性;Westernblot检测表明MG63细胞MMP-2和MMP-9的表达明显下调。结论 NDGA可抑制骨肉瘤MG63细胞的体外黏附,迁移和侵袭,这种抑制效应可能是通过减少骨肿瘤细胞MMP-2和MMP-9的表达来实现。Objective To study the effects and its potential molecular mechanism of nordihydroguaiaretic acid on adhesion, migration and invasion of osteosarcoma MG63 cell line. Methods Adhesion, migration and invasion of nordihydroguaiaretic acid MG63 cells treated by were measured by CCK-8 assay, wound assay and Transwell chamber method respectively. Western blot analysis was used to evaluate the expression of MMPs protein. Results Nordihydroguaiaretic acid inhibited the adhesion, migration and invasion of MG63 cells in a dose-depended manner (P 〈 0. 05 ). The protein levels of MMP-2 and MMP-9 were down-regulated. Conclusion Nordihydroguaiaretic acid has anti-adhesion, anti-migration and anti-invasion effects on osteosareoma MG63 cells through down-regulating the levels of MMP-2 and MMP-9 in vitro.
