Balb/C鼠柯萨奇病毒B_3心肌炎模型中病毒接种浓度的研究

摘　　要：选取 ( 4～ 6)周龄雄性 Balb/ C小鼠 ,病毒感染组腹腔接种不同浓度的 CVB3病毒液 ,对照组注射 RPMI1 640液 ,7d后测定血清中和抗体效价及乳酸脱氢酶 ( LDH)活性 ;测定心肌组织的病毒滴度及观察心肌组织病理学改变。结果显示 :( 1 )病毒感染组鼠血清中和抗体效价及乳酸脱氢酶活性显著高于正常对照组 ,且随接种 CVB3浓度增加 ,L DH活性增高。( 2 )随接种 CVB3浓度增高 ,小鼠心肌组织病毒滴度增加不明显 ;( 3)病毒感染组心肌组织出现不同程度的灶性病理改变。 ( 4 )接种病毒浓度与鼠心肌炎发病率之间不存在剂量反应关系。Male Balb/C mice (4～6 weeks old) were selected.Mice of CVB_3-infected groups were inoculated intraperitoneally with different concentration of CVB_3 and mice of control group were inoculated with sterile RPMI1640.At day 7 postinfection blood specimens were detected for neutralization antibody and lactate dehydrogenase (LDH).The heart specimens were titrated for 50 % tissue culture infection dose (TCID_ 50 ) and were examined for histological lesions.The results showed:1.Serum neutralization antibody titers and LDH activenesses of mice in CVB_3-infected groups were significantly higher than those of mice in control group and this effect was dose dependent for LDH.2.CVB_3 titers of myocardial tissue in CVB_3-infected groups were not significantly increased with viral concentrations inoculated increasing.3.There were varying extent of focal histological lesions of hearts in CVB_3-infected groups.4.There was not dose reaction relationship between morbidity rate of myocarditis and concentration of CVB_3 inoculated.
