咖啡因对小鼠脑内纹状体CART肽表达的影响研究

摘  要:目的:研究咖啡因对小鼠脑内纹状体可卡因安非他明调节转录(CART)肽表达的影响。方法:取小鼠随机分为对照组(生理盐水)和低、中、高剂量咖啡因组(10、30、100mg·kg-1),每组10只,灌胃给予相应药物,每日1次,连续7d,考察给药前后各组小鼠运动活性,以产生行为敏感化为指标选取最佳给药剂量和时间;再考察最佳给药时间不同剂量咖啡因组脑内纹状体中CARTmRNA及其蛋白水平,以及最佳给药剂量不同给药时间脑内纹状体中CARTmRNA及其蛋白水平。结果:与对照组比较,各剂量组小鼠运动活性明显增加(P<0.01和P<0.001);与各剂量组第1天比较,中剂量咖啡因组小鼠在第5天时运动活性明显增加(P<0.05),表明产生行为敏感化。与对照组比较,中剂量咖啡因组纹状体中CARTmRNA水平在第3、5天时明显增加(P<0.05或P<0.01)、CART蛋白水平在第5天时明显增加(P<0.05),并且第3天CARTmRNA水平达峰值,第5天CART蛋白水平达峰值。结论:小鼠脑内纹状体中CART肽是咖啡因行为敏感化产生的重要调节因子。OBJECTIVE: To study the effects of caffeine on CART peptide expression in cerebral striatum of mice. METH- ODS: Mice were randomized into control group (normal saline) and caffeine low-dose, medium-dose and high-dose groups (10, 30, 100 mg.kg^-1) with 10 mice in each group. They were given relevant medicine once a day for consecutive 7 d. The locomotor activity of mice was investigated before and after medication, and the best dosage and best medication time were screened by using sensitization as index. Subsequently, mRNA and protein expressions of CART in cerebral striatum were investigated in different dos- es of caffeine groups at the best time point, and those of CART in cerebral striatum were also investigated in different time groups with the best dose. RESULTS: Compared with control group, the locomotor activity of mice increased significantly (P〈0.01 or P〈0.001) ; compared with the first day, that of mice in medium-dose caffeine group increased significantly at the fifth day (P〈 0.05), indicating behavioral sensitization. Compared with control group, the mRNA expression of CART in medium-dose caffeine group increased significantly at the third and fifth day (P〈0.05 or P〈0.01) ; the protein expression of CART increased significant- ly at the fifth day (P〈0.05) ; the mRNA expression of CART reached the peak at the third day and the protein expression of CART reached the peak at the fifth day. CONCLUSIONS: CART peptide is necessary in the process of behavioral sensitization in cerebral striatum of mice by caffeine.
