摘  要:目的筛选肠道干细胞特征性的差异蛋白质或蛋白质群。方法采用双向凝胶电泳方法分离新生小鼠肠边缘群细胞与肠上皮细胞总蛋白,表达差异超过1.5倍的为差异蛋白质。结果成功建立小鼠肠边缘群细胞与肠上皮细胞的双向电泳图谱,筛选出差异蛋白点39个。结论小鼠肠干细胞与上皮细胞的蛋白表达谱存在差异,有可能成为潜在的、有应用价值的肠道干细胞鉴别分子标志。Objective It is to screen differential proteins or protein groups with stem cell characteristics in intestinal stem cells. Methods Two-dimensional gel electrophoresis was used to separate total proteins from intestinal side population cells and intestinal epithelial cells of neonatal mice. Proteins which were differentially expressed more than 1.5 times were selected as differential proteins. Results The two-dimensional electrophoresis patterns of intestinal side population cells and intestinal epithelial cells were successfully established to screen 39 differential protein spots. Conclusion There were differences between the protein expression profiles of mouse intestinal epithelial cells and stem cells. The proteins may become potential and valuable molecular markers for identification of intestinal stem cells.
